Objective To explore the epileptogenesis effect of uPA and uPAR in the rat cerebral cortex with cortical dysplasia(CD) byγ-ray irradiated injury. Methodsγ-ray induced CD model rats were established and randomly divided into different groups according to postnatal age (neonatal rats, 2 week, 4 week, and adult rats) . The pathologic examination was performed, including hematoxylin and eosin(HE), immunohistochemistry and immunofluorescence. We observed the pathological change and the expression of uPA and uPAR in the rat cerebral cortex under a light microscope. Results Immunohistochemistry showed uPA-positive and uPAR-positive cells in each group. Along with the growth of the age, uPA-positive cells obviously increase, uPAR-positive cells obviously decrease. Compared with the normal control group, the expression of uPA was lower in testis of model groups,the expression of uPAR was higher. Double-labeled immunofluorescence staining showed that uPA-positive and uPAR-positive cells were mainly neurons. Conclusion By ray-induced CD model, this study confirms the CD rats with age, uPA-positive cells significantly decreased and uPAR positive cells significantly increased, and mainly expressed in neurons. In the process of CD, uPA and uPAR may play an important role in the neural organization migration. These results provided theoretical and technical support for research uPA and uPAR in the mechanism of epileptogenesis.
