| AIM: To investigate the expression of Melanoma antigen-1(MAGE-1)and New York-esopHageal-1(NY-ESO-1)mRNA in the tissues of gastrointestinal stromal tumor(sGISTs),to discuss the possibility and the clinical significance of MAGE-1 and NY-ESO-1 antigens used as specific targets for the immunotherapy of GISTs,and to evaluate the possibility of MAGE-1 and NY-ESO-1mRNA used as auxiliary grading- criteria for GISTs dangerousness.Methods: MAGE-1 and NY-ESO-1mRNA were detected by reverse-transcriptase polymerase chain reaction in tissue specimens of tumor in 30 patients with GISTs.The pathological data were retrospectively analyzed.Results:The rate of MAGE-1 and NY-ESO-1mRNA expression was 33.4%and 46.6% respectively in tumor tissues of GISTs ,18 cases of GISTs tissues were detected at least one of these two CTAs.The expression of MAGE-1 and NY-ESO-1mRNA was unrelated to age,sex or pathology type(P>0.05),but related to the original site,tumor diameter and the biological potential grading(P<0.05).The more dangerous it becomes, the more MAGE-1 and NY-ESO-1mRNA expressed,there was significant difference in MAGE-1 expression and NY-ESO-1mRNA(P<0.05).The expression of MAGE-1 and NY-ESO-1mRNA in GISTs were negatively correlated(r=0.189,P>0.05).Conclusion:(1) MAGE-1 and NY-ESO-1mRNA were expressed with a high specificity in the tissues of GISTs,thus their antigens might be potentially promising targets for antigen-specific immunotherapy of GISTs.(2) The expression of MAGE-1 and NY-ESO-1mRNA were correlated with the grading-criteria of GISTs dangerousness. MAGE-1 and NY-ESO-1mRNA are the promising auxiliary index to diagnose the GISTs dangerousness. |
