Objective:Xifeng zhitong oral liquid on the threshold dose of sodium pentobarbital-induced sleep were studied in mice, on the threshold dose of pentobarbital sodium in mice induced sleep time and sleep latency effects; Xifeng zhitong oral liquid chemical experiments in mice induced by hypoxia effects; of Xifeng Liquid on migraine pain model rats the blood of 5 - hydroxytryptamine (5-HT) andβ-endorphin (β-EP) and the brain stem of nitric oxide (NO) effect. And to explore its possible mechanism of action, clinical development for the prevention and treatment of migraine provide experimental basis for Chinese medicine.Methods:The literature commonly used modeling approaches, the following experiment: 1. Subthreshold dose of sodium pentobarbital on mice of sleep rate and the threshold dose of pentobarbital sodium on sleep latency and mice the impact of sleep time. Application of the threshold dose of pentobarbital sodium (35mg/kg) and the threshold dose (55mg/kg), testing whether the drug sodium pentobarbital synergistic role in enhancing the rate of infected mice of sleep, to extend the subjects mice reduced sleep time and sleep latency, which can show that the drug has a sedative effect to relieve pain.2. On chemical induced hypoxia experiment. To infected mice by intraperitoneal injection of sodium nitrite in mice, the survival time after injection, testing of the drug could prolong anoxic survival time of mice. 3. Nitroglycerin rat model of migraine in bloodβ-EP and 5-HT content. Modeling using nitroglycerin, get migraine model, the rats were decapitated to collect mixed venous blood, the measuredβ-EP and 5-HT values testing the drug on serumβ-EP and 5-HT whether the impact of the content. 4. Nitroglycerin in rats with NO content in the brain stem. Application of nitroglycerin create animal models of migraine, After the modeling, the rats were decapitated and quickly remove the rodent brain, isolated brain stem part, weighing with saline ground, made into suspension, and then the supernatant NO content in brain stem testing, testing the drug on the rat brain stem in NO content.Results:1. The experimental results show that high dose and medium dose of Xifeng zhitong oral liquid can significantly increase the threshold dose of sodium pentobarbital induced sleep ratio (P<0.05), but in the low dose had no significant effect. Xifeng zhitong oral liquid high dose and medium doses can significantly prolong the sleeping time (P<0.05), its high and low doses significantly shortened sleep latency (P<0.05). 2. Xifeng high, medium and low doses tested in mice could significantly prolong the survival time under hypoxic conditions (P<0.05). 3. Xifeng high, medium and low dose significantly higher bloodβ-EP content and 5-HT content (P<0.05). 4. Xifeng zhitong oral liquid can significantly reduce the NO content in rat brain stem (P<0.05).Conclusion:Xifeng zhitong oral liquid with high doses of oral sedation, with hypoxia tolerance; Xifeng zhitong oral liquid can increase model of oralβ-EP in blood levels and lower NO levels, in order ease the headache; Xifeng zhitong oral liquid can increase model in the rat brain stem 5-HT levels, reduce pain.
