Part 1 Theoretical studyBy reviewing and summing up:TCM literature on insulin resistance (IR) related diseases Pathogenesis, TCM pathological features, Include:the relationship between the kidney deficiency and the IR, the relationship between the Spleen deficiency and the IR, the relationship between phlegm obstruct and the IR, the relationship between the blood stasis and the IR, TCM nourish kidney law, strengthen spleen law, circulate the blood and resolve stasis law, resolve phlegm law treatment effects, as well as IR animal model application experience, we have come to the basic TCM pathological changes of IR is the Ben Deficiency Biao Excess and Interlocking of Deficiency-Excess. The Deficiency Syndrome were mainly spleen deficiency, kidney deficiency, the Excess Syndrome were mainly phlegm and blood stasis. Dialectical treatment of TCM of IR should be from the kidney deficiency, spleen deficiency, phlegm obstruct and blood stasis to start. Preparation of animal model of IR to simple diet induced a more economical and practical, and this model is more consistent with the man who may because of modern living standards, often "too fresh Feigan," over-nutrition, and thus causing the basic pathogenesis features of IR.Part 2 Experimental studyObjective:This object to be established IR rat model; to explore different results of Chinese herbal compound interfere with IR rat models, disproof the TCM pathological features of IR rat model; to explore the Correlation between IR rat model and" TCM Syndrome"; to identify the hidden relationship among the FPQ FINS, TG, TC changes in IR animal models, the liver, spleen, kidney visceral pathological changes, and the "TCM Syndrome", hope in the future for the IR Diseases to establish a targeted, the system of Chinese medicine prevention and control measures, for the future development of effective prevention and treatment of drug delivery a meaningful reference.Methods:68 healthy male Wistar rats were randomly assigned into 2 groups:normal diet group,8, and high-lipid diet model group,60, were given ordinary feed and high-lipid feed respectively.1 month later, High-lipid diet model group were re-randomized into high lipid control group, high-lipid kidney tonic group, high-lipid spleen strengthening group, high-lipid phlegm resolving group and high-lipid stasis elimination group 5 groups. Then, right outside of the high lipid control group rats, in each group were used to Liu Wei Di Huang Tang, Si Jun Zi Tang, Er Chen Tang, Xue Fu Zhu Yu Tang intervention of four kinds of Chinese herbal compound. After 2 weeks of TCM differential intervention, kill rats, take body weight before and after modeling, as well as the appearance and behavior; discover FPG, TG, TC, FINS; observe pathological changes of the liver, kidney, spleen.Results:After modeling,1.high-lipid diet model group rats and normal rats have a very significant body weight increased (P<0.01), however, compared two groups of rats, body weight was no significant difference (P> 0.05); 2.there were Spleen Deficiency Syndrome, Blood Stasis Syndrome, and some of Kidney Deficiency Syndrome performances of TCM in High-lipid diet model group rats'appearance and behaviors; 3.high-lipid diet model group, the insulin sensitivity index (ISI) lower than the normal control group, the natural right Number of insulin sensitivity index (IAI) of the statistical analysis showed that the former values were significantly lower (P<0.01); 4.FPG and FINS levels of all groups of high-lipid diet model group rats compared with normal control group were significantly higher (P<0.01).Chinese medicine intervention 2 weeks later, l.in FPG, high lipid control group 's significantly higher than the normal control group (P<0.05), high-lipid stasis elimination group also significantly increased (P<0.01), high-lipid kidney tonic group, high-lipid spleen strengthening group, high-lipid phlegm resolving group and normal control group showed no significant difference (P> 0.05); but compared with the high lipid control group, the FPG of these three groups were significantly lower (P<0.05); 2.in FINS, compared with the normal diet group, high lipid control group's FINS level is still significantly higher (P<0.01), high-lipid stasis elimination group rats compared with normal control group had significantly lower levels (P<0.05), other groups and normal control group, no significant difference between, however, compared with the high lipid control group, after intervention of traditional Chinese medicine, in each high-lipid diet model group, the levels of FINS significantly lower (P<0.01); 3.in TG, compared with the normal diet group, high lipid control group significantly increased (P<0.05), high-lipid phlegm resolving group and high-lipid stasis elimination group than in the normal control group were decreased more significantly (P<0.01), high-lipid spleen strengthening group significantly increased (P <0.05); compared with the high lipid control group, high-lipid kidney tonic group significantly lower (P<0.05), high-lipid spleen strengthening group significantly increased (P <0.05), high-lipid phlegm resolving group and high-lipid stasis elimination group were decreased more significantly (P<0.01); 4.in TC, compared with the normal diet group, high-lipid phlegm resolving group significantly decreased (P<0.01), other groups and normal control group, no significant difference between; compared with the high lipid control group, high-lipid kidney tonic group, high-lipid spleen strengthening group, high-lipid stasis elimination group, all decreased significantly (P<0.05) significantly increased (P<0.05), high-lipid phlegm resolving group significantly decreased (P<0.01).In the pathological aspects,1.pathological changes in the liver are:compared with the normal diet group, high lipid control group's liver cells fatty degeneration obviously, fatty degeneration of high-lipid kidney tonic group obvious, fatty degeneration of high-lipid spleen strengthening group obvious, high-lipid phlegm resolving group fatty degeneration of liver cells, and liver cells with mild edema, high-lipid stasis elimination group mild fatty degeneration of liver cells, in each group was no other pathological changes in liver tissue; 2.main pathological changes of kidney are:compared with the normal diet group, high-lipid kidney tonic group mild bleeding, mild cloudy swelling of renal tubular, high-lipid stasis elimination group mild bleeding, mild cloudy swelling of renal tubular, the other groups were seen in stasis blood or cloudy swelling; 3.spleen of rats in each group were no obvious abnormalities.Conclusion:1.prepared by high-lipid diet-induced IR rat model may be mainly closely related to TCM Phlegm Obstruct Syndrome, Blood Stasis Syndrome, and There is some relevance to both Kidney Deficiency Syndrome and Spleen Deficiency Syndrome.The TCM pathological features of IR rat model are:mainly phlegm obstruct and blood stasis, both kidney and spleen deficiency, Interlocking of Deficiency-Excess, mainly Excess Syndrome.2. FPG increased may be phlegm blocked, both kidney, spleen deficiency results from the cooperation; FINS elevated may be blood stasis, accompany by block phlegm, kidney deficiency, and the joint effect of spleen deficiency; the increase of TG may be mainly affect in combination with phlegm obstruct and blood stasis, both kidney deficiency; the increase of TC may correspond to the TCM pathological features of mainly block phlegm, accompany by blood stasis, kidney deficiency, spleen deficiency. This shows that biochemical indicators of Western medicine and TCM between the pathological features is not a simple one to one relationship.3.The pathological changes of the liver, the kidney of IR rat model, may closely related to Phlegm Obstruct Syndrome, Blood Stasis Syndrome.
