Study On The Effects Of Inhibiting Of Pomegranate Husk Extract On Human Breast Cancer Cell

Composed of two parts which include theoretical and experimental research,it discus about the Effect and its Mechanisms of Pomegranate husk extract on inhibiting Proliferation of the hu-man breast cancer cell.Theoretical study involves the clinical reach of Western medicine and Chinese medicine on the breast cancer in recent years.On this basis,We synthesized the clinical experience of instruct-ors and academic thinking and discussed the the evidence of "JieduSanjie"(detoxification and dispersing accumulation).Expounded the theory that"Aidu"(poisonous pathogenic factor) is the etiological factor and pathologic product through the whole course of breast cancer."TanduYujie "(stagnation of phlegm,poison and blood stasis) is the essential pathogenesis of the breast cancer 's development."YuduPangcuan"(vestigial poison invasion elsewhere) is the key pathogenesis of recurrence and metastasis after operation."JieduSanjie"(detoxification and dispersing accumulat-ion) is an important therapeutic principle in breast cancer's treatment.Ancient literature recorded that the pomegranate has detoxification effect.Modern medicine confirms that pomegranate can inhibit human breast cancer cells.To study the condition and mechanism of action of treatment of breast cancer induced by Pomegranate husk extract,we conducted a number of experiments.Methods:(1) The MTT assay was used to detect the growth inhibition rates of MCF-7 cells and MDA-MB-231 cells at 24,48, 72h which cultured with Pomegranate husk extract in different concentrations.(2) Flow cytomet-ry, AnnexinV/PI double staining method was used to observe the apoptosis and the cell cycle dis-tribution induced by Pomegranate husk extract in human breast cancer cell line MDA-MB-231. Results:(1) Pomegranate husk extract inhibited the proliferation of human breast cancer cell line MCF-7 and MDA-MB-231.The different concentrations of Pomegranate husk extract had cytoto-xicity on breast cancer cells, whichwere dose and time-dependent. (2)Cells were treated with Po-megranate husk extract which had effects on cell cycle of breast cancer cell line MDA-MB-231 and induced apoptosis,whichwere time-dependent..Cell cycle was arrested in G2 phase.Conclusi-ons:Pomegranate husk extract inhibit human breast cancer cell line MCF-7,MDA-MB-231 cell proliferation, and can induce breast cancer cell line MDA-MB-231 apoptosis and affect the distribution of the cell cycle.
