AIM:To determine whether the lentivirus mediate Light gene can inhibit the HCT116 cells.METHODS:Using lentivirus mediate Light gene infect HCT116 cells.The cells were devided into three groups.In order to find out the changes of proliferation of HCT116 cell lines and the expression of Light gene mRNA in HCT116 cell lines. We analyzed the data by real-time fluorescent quantitative PCR and MTT experiment.RESULTS:1.The experiment of real-time fluorescent quantitative PCR showed the positive expression of Light gene mRNA(23.75243±1.16197)was stronger than the negative group(2.10372±1.26214)and control group.P<0.052.The values of 490 nm wavelength light absorption were different in the three groups in MTT experiment. The number of alive cells in the group of Light was lower than others in the frist day(P< 0.05). The number of alive cells in the negative group was lower than control group from the second day(P<0.05).2CONCLUSION:Light genes mediated by lentivirus may be useful for inhibiting growth of HCT116.These results provide a novel therapeutic preventing colon cancer cell invasion and metastasis.
