Objective To constructe overexpression of mIL12mediated by lentivirus,foster the mIL12-BMSCs,which make mIL12gene stable transfection and high expression in the cells,andexplore its effect on colorectal cancer (CT26) in rats.Methods In this study,we designed and synthesized primers of p35and p40,amplified themby PCR,constructed mIL12fusion gene by Overlapping PCR,and transfected it toBMSCs,then collected and fostered the mIL12-BMSCs.The rat models were divided into4groups.Rats were injected peritumoral once every4days in12days,group A were injected ofmIL12-BMSCs(0.2ml, approximately2×106cells), group B PBS(0.2ml), group C BMSCs(0.2ml, approximately2×106cells),group D rLV/IL12(0.2ml, approximately2×109virus).Atlast,we statisticed and analysised the data of tumor volume.Results We found that the data showed no significant difference between group A and D,aswell as group B and C(P﹥0.05).However,from the7thday,data of group A and B, B and Dshowed a significant difference(P﹤0.01).From the10thday, data of group A and D alsoshowed a significant difference(P﹤0.05).Conclusion We demonstrated that mIL12-BMSCs could inhibit the growth of CT26tumorsignificantly.
