AIM:To evaluate the effect of the lentivirus mediate human TNF-βgene on the colorectal cancer cells HCT116 cells.METHODS:Cultured HCT116 cell line.Using lentivirus mediate TNF-βgene infect HCT116 cells.The cells were devided into three groups.The cells were infected with lentiviral vector-mediated TNF-βgene in cells (experimental group), no TNF-βgene lentiviral cell group (negative control group), HCT116 cells (control group). In order to find out the changes of proliferation and mRNA and proteinum level of HCT116 cell lines, we analyzed the data by real-time fluorescent quantitative PCR and MTTexperiment.RESULTS:HCT116 cells of the expression of green fluorescent protein was found in cells infected with lentivirus 48h, and cells infected with efficiency greater more than 95% from observed by fluorescence microscopy,so,slow lentivirus infectied HCT116 cells efficiency was very highly. Real-time quantitative RT-PCR date showed that lentiviral vector-mediated TNF-βgene infected on HCT116 cells the TNF-βmRNA at 12h,24h,48h,72h, and 5d were significantly higher than the negative control group and control group, and the difference was significant (P<0.05), while the negative control group and control group was no significant differenct (P>0.05). The values of 490 nm wavelength light absorption were different in the three groups in MTT experiment. The number of alive cells in the group of TNF-βwas lower than others in the frist day (P< 0.05). While the negative control group and the control group no difference (P>0.05).The number of alive cells in the negative control group was lower than control group from the second day,and the difference was significant(P<0.05).It showed HCT116 growth slow down infected by lentivirus.CONCLUSION:TNF-P genes mediated by lentivirus may be useful for inhibiting growth of colorectal cancer HCT116. These results provide a new therapeutic preventing colorectal cancer.
