Objectives: Glomerular nephritis is an allergic disorder caused by multireasons induced glomerular damage.Many experiments and clinical study shows that glomerular nephritis was a immunologic disease caused by antigen-antibody reaction.As the disease incidence of glomerular nephritis raises year by year,the therapeutic tools of glomerular nephritis were paid more and more attention.The drugs used now in clinically was immunodepressant and excitatory autacoid,which have many drug side effects. So it's hot spot to find more efective and safer drugs to cure glomerular nephritis. Traditional Chinese medicine consider that Cordyceps could invigorate the kidney and tonifying kidney, and to balance yin-yang. Based on the early research of our laboratory, we continue to study the pharmacological activity of different extractive of Cordyceps to inspect that if it is could to develop as the therapeutic drug of glomerular nephritis.Methods: This study use in vivo and in vitro experiment to research the impact on immunologic system and therapeutical effect of glomerular nephritis of the extractive of Cordyceps.①In vitro, including the impact on immunologic system of the extractive of Cordyceps and the dosage range. And to observe the effective of extractive of cordyceps on glomerular mesangial cell proliferation, lymphocyte transformation in mice, peritoneal macrophage phagocytosis,to research the impact on immunologic system;②In vivo,through the experiments of carbon clearance in mice and delayed-type hypersensitivity in mice,we study the the overall impact on the immune system to confirm its function and to lay the foundation for follow-up study.③Construct the anti-thymocyte serum nephritis model to study the therapeutical effect on Mesangial Proliferative Glomerulonephritis of n-butanol extract of Cordyceps.By measuring the 24h urinary protein of animal model, content of circulating immune complexes, erythrocyte rosette and Splenic lymphocyte transformation function,we analysis the pharmacological effects of n-butanol extract of Cordyceps.In additional, we use electron microscope and PAS staining biopsy to obverse the success of animal model making,and to inspect the preventive and therapeutic effect of n-butanol extract of Cordyceps to anti-thymocyte serum nephritis model。 Conclusion:1,Impact on glomerular mesangial cell proliferation: Methanol, n-butanol and ethyl acetate extracts of Cordyceps could inhibit the proliferation of normal mesangial cells, but the Methanol and n-butanol extracts of Cordyceps were more sensitive to mesangial cells stimulated and actived by LPS.2, The impact of immune cells: Ethyl acetate and n-butanol extracts of Cordyceps could significantly inhibit the transformation of lymphocyte,and in high concentration the two extracts could inhibit the phagocytosis of peritoneal macrophage. Separation of part of 70% ethanol which was Separated from n-butanol extracts by macroporous resin,could inhibit the phagocytosis of peritoneal macrophage in high concentration and to promote the phagocytosis of peritoneal macrophage when low concentration.3,The clear of circulating immune complexes: N-butanol extract 10mg/kg treatment group and n-butanol extract 20mg/kg treatment group could cut down the level of circulating immune complexes of model animals and the rate of CIC erythrocyte rosette.The 24h urinary protein of two treatment groups was reduced, and the 10mg/kg treatment group was more effective than 20mg/kg treatment group.4,The impact on anti-thymocyte serum nephritis model of n-butanol extract: Biopsy(PAS, Masson staining and electron microscope) show that in model rats, mesangial matrix was dissolved,mesangial cell proliferation, basement membrane thickening, foot process fusion and Some glomerular was necrosis. Pathological indicators of two treatment groups were improved,and the 10mg/kg treatment group was more effective than 20mg/kg treatment group.Result:The different extracts of Cordyceps has different impact on immune system, the thyl acetate extracts could inhibite the proliferation of mesangial cells, transformation of lymphocyte,and the phagocytosis of peritoneal macrophage.The n-butanol extract main shows concentration-related immune suppression or immune regulation on these experiments and delayed-type hypersensitivity in miceIn vivo experiments show that n-butanol extract of Cordyceps could improve the indicators of the pathology in rat model of glomerulonephritis, it's prove that n-butanol extract has the value of further study in glomerulonephritis prevention and therapy...
