Cordyceps militaris has biological activities and high medicinal value because of a variety of nutrients. Cordyceps polysaccharides is the main active ingredient. Metabolomics,a new subject, is aimed to analyze the small molecular (M<1000) metabolites produced in an organism or cell. Through the analysis of metabolites and comparison with the normal metabolism, biomarkers of diseases will be identified. It can provide a way for disease diagnosis. In this study, the hypoglycemic activity and immune activity of Cordyceps militaris by means of metabonomics were explored to find some potential biomarkers. The main research methods and results are as follows:Polysaccharides from Cordyceps militaris were extracted by water extraction, alcohol precipitation and enzymatic removal of protein. The yield of polysaccharides was 5.36%.The content of soluble sugar of polysaccharides was 56.2% by the analysis of Phenol sulfuric acid method.STZ-induced hyperglycemia model mice were used to test the hypoglycemic activity of Cordyceps polysaccharides. The metabolic profiling method -was adopted to find the related biomarkers and the metabolic pathway of diabetes. Results showed that 120 mg/kg Cordyceps polysaccharides can effectively reduce the blood glucose of diabetic mice, and regulate the body's energy metabolism, amino acid metabolism and intestinal microbes.Biomarkers are glucose, lactic acid, 3-hydroxy butyric acid, creatine, glutamate, valine,leucine, isoleucine and VLDL.Immunosuppressive model mice induced by cyclophosphamide were applied to verified the immune activity of Cordyceps polysaccharides. The metabolic profiling method was used to find the associated biomarkers and the metabolic pathway of immune suppression. Results showed that 200 mg/kg Cordyceps polysaccharides can significantly improve the immunity of -the immune suppression mice. High-dosage Cordyceps polysaccharides can significantly increase the immune organ weight and the number of blood cells and the content of three kinds of immunoglobulin. The metabolism of immunosuppressive model group caused by cyclophophamide was in disorder. Catabolism of the body reduced, whereas, anabolism strengthen. Some related potential biomarkers were glucose, lactic acid, 3-hydroxy butyric acid, pyruvate, glycoprotein and glutamine.Cordyceps polysaccharides can improve the metabolic status and immunity of the immunosuppressive model group through regulating the internal environment and immune organs.
