Objective:To determine the expression of high mobility group protein boxl(HMGB1) in endometrial stromal sarcoma, and to investigate the role of HMGB1 in the pathogenesis of endometrial stromal sarcoma. Methods:Immunohistochemical method was employed to detect the expression of HMGB1 in endometrial stromal sarcoma, TUNEL method was employed to detect the level of apoptosis in endometrial stromal sarcoma.Results:1, HMGB1 in endometrial stromal sarcoma was mainly localized in the nucleus and in normal endometrium was only localized in the nucleus. No significant expression in the cytoplasm was found in normal endometrium. The positive expression rate of HMGB1 in endometrial stromal sarcoma were significantly higher than that in normal endometrium (P<0.05);2, The level of apoptosis in the endometrial stromal sarcoma was significantly lower than that in the nomal endometrial (P<0.05);3, The positive expression rate of HMGB1 expression in endometrial stromal sarcoma was associated with the degree of malignancy, stage, myometrial invasion, and lymph node metastasis, but independing on menopause, age.4, In the endometrial stromal sarcoma the apoptosis level of the cells expressing HMGB1 was lower than that in the cells without expressing HMGB1. (P<0.05).Conclusion:1, HMGB1 may play a role in the pathogenesis, invasion and metastasisbe of endometrial stromal sarcoma,by its biological effect is working by its moving from nucleus to cytoplasm.2, HMGB1 may play a role in the pathogenesis of endometrial stromal sarcoma,by inhibitting the process of apoptosis of tumor cells.3, HMGB1 may play a role in the invasion and metastasisbe of the endometrial stromal sarcoma.
