| Objective:To detect the high mobility group protein 1(HMGB1)gene expression level in the peripheral blood of children with Henoch-Schonlein purpura(HSP),analyze its correlation with laboratory indicators,and preliminarily explore the role of HMGB1 in the pathogenesis of HSP,for the future The clinical treatment of HSP provides a theoretical basis.Method : Real-time PCR was used to detect the relative expression of HMGB1 m RNA in peripheral blood of 21 children with Henoch-Schonlein purpura(HSP group),16 children with purpura nephritis(HSPN group)and 22 cases of control group children,and detect the relative expression of HMGB1 m RNA in the peripheral blood of HSP group and HSPN group The child’s serum white blood cell count(WBC),percentage of neutrophils(NE),C-reactive protein(CRP),interleukin-6(IL-6),tumor necrosis factor-a(TNF-a),platelets(PLT),Laboratory indicators such as D-dimer and immunoglobulin A(Ig A).Result:(1)The relative expression of HMGB1 m RNA in peripheral blood of children in HSPN group,HSP group and control group was 15.88(11.54~61.34),9.65(3.80~26.72),1.01(1.00~1.06),respectively.The expression of HMGB1 m RNA in HSPN group was higher than In the HSP group and the control group,the expression level of the HSP group was higher than that of the control group,and the difference was statistically significant(P < 0.05);(2)The peripheral blood HMGB1 m RNA of the mixed type,abdominal type and simple type in the HSP group The expression levels were 18.14(10.54~28.97),12.19(4.93~32.60),2.93(0.88~5.69),among which the expression levels of children with mixed and abdominal HSP were higher than those with simple HSP,and the difference was statistically significant(P<0.05),the expression level of mixed type children was higher than that of abdominal type children,but the difference was not statistically significant(P>0.05);(3)Peripheral blood HMGB1 m RNA expression and WBC(r=0.89),NE(r=0.93),CRP(r=0.73),IL-6(r=0.82),TNF-a(r=0.48),PLT(r=0.78),D-dimer(r=0.59),Ig A(r=0.64)and other indicators were positively correlated,all P<0.05.Conclusion:The expression of HMGB1 m RNA in the peripheral blood of the HSPN group and HSP group was significantly higher than that of the control group,indicating that HMGB1 may be involved in the pathogenesis of HSP and is related to renal damage;the expression of HMGB1 m RNA in children with mixed and abdominal types in the HSP group was significant Higher than simple children,it is speculated that HMGB1 can be used as an indicator of the severity of HSP disease;the test group’s peripheral blood HMGB1 is positively correlated with serum WBC,NE,CRP,IL-6,TNF-a,PLT,D-dimer,etc.It is speculated that HMGB1 is involved in disease inflammation and hypercoagulable state. |
