Objects:To research the expression of WIF-1 (wnt inhibitory factor-1) in human brain glioma tissue and to research the relationship with the pathological grade and with the patient age group and gender.Methods:43 cases of fresh human glioma specimens and 4 cases of normal brain tissue specimens was collected from Neurosurgery in Xiangya Hospital,Central South University in 2008 -2009. The relative WIF-1 mRNA expression of all these specimens was calculate respectively, by RT-PCR, and agarose gel electrophoresis, again housekeeping gene comparison. Finally they are tested by the statistical analysis whether the WIF-1 mRNA expression in glioma cells is significantly lower than the one which is in normal brain cells, and whether WIF-1 mRNA expression level has relationship with the pathological grade and with the patient age group and gender. All of these specimens was made into paraffin sections,.respectively, WIF-1 protein immunization points was calculated respectively by SP immunohistochemistry staining, refering to Joki immune response integration method (IRS). Finally they are tested by the statistical analysis whether the WIF-1 protein expression in glioma cells is significantly lower than the one in normal brain cells, and whether WIF-1 protein expression level has relationship with the pathological grade or with the patient age and gender.Results:The WIF-1 mRNA relative expression and WIF-1 protein immunization points in glioma cells are both significantly lower than the ones in normal brain cells;The ones in high-grade glioma cells are significantly lower than the ones in low-grade glioma cells.But The ones are not significantly different between different patient age group or different gender.Conclusion:1. The WIF-1 expression in glioma cells is significantly lower than the one in normal brain cells in both mRNA and protein levels.2. The WIF-1 expression in high-grade glioma cells is significantly lower than the one in low-grade glioma cells. 3. The WIF-1 expression has no significant correlation with the patient age and gender.4. The WIF-1 gene may be the suppressor gene of glioma, may be one of its new reference of pathological grade-determining and prognosis-evaluating and may be its new targets to therapy.
