Objective:To study the mechanism of OPN in Viral Myocarditis by observing the expression of OPN in mice myocardium.Methods:Make the Viral Myocarditis model with BALB/C mice infected by CVB3 (n=32),and set the control group (n=32),get the myocardial and do HE staining and Masson staining, account the pathological scores and the CVF of myocardium. Detect the content of Colâ… by RT-PCR method, and detect the expression of OPN by RT-PCR and ELISA method, and carry out correlative analysis.Result:1. Coxsackie virus B3 can used to establish the viral myocarditis model of BALB/C mice successfully.2. The histopathological examination revealed that lots of myocardial cell necrosis occurred, found obvious inflammation changes in acute stage of VMC, then the inflammatory cells had almost disappeared and took on the obvious fibrosis in recovery stage.3. The pathological scores of myocardium:The pathological scores in VMC group were high than that in control group (P<0.05), it obviously increased in the 7th day, then gradually decreased from the 14th day, and it reduced to minimum in 28th day.4. Detect the expression of OPN by RT-PCR:There were expression of OPN mRNA in normal and VMC group's myocardium, the OPN expression reached the highest at 7th day then its activity decreased sharply from 14th day, then it reduced to minimum in 28th day in VMC group, but which all high than that in control group(P<0.05).5. Detect the expression of OPN by ELISA method:The concentration of OPN in VMC group reached the highest at 7th day then its activity decreased sharply from 14th day, when it reduced to minimum in 28th day, which all high than that in control group(P<0.05).6. The result of CVF showed:The CVF at 7th day and 14th day in VMC group were no obvious differences compared with control group (P>0.05); the CVF in 21th day and 28th day in VMC group had obvious difference compared with that in control group,7th day and 14th day in VMC group(P<0.05). 7. Detect the expression of Col I mRNA by RT-PCR and ELISA method: The Col I mRNA expression were weak at 7th day and 14th day in VMC group, there were no obvious differences compared with control group (P>0.05), then its enhanced gradually from 14th day, and it reached the highest in 28th day, the OD value of Col I mRNA/GAPDH in 21th day and 28th day in VMC group had obvious difference compared with that in control group,7th day and 14th day in VMC group(P<0.05).8. The correlative analysis of OPN expression and the pathological scores of myocardium, R2=0.8877.They have positive correlation; the correlative analysis of Col I mRNA expression and the CVF, R2=0.9411.They have positive correlation. The correlative analysis of Col I mRNA expression and the OPN mRNA expression,R2=0.3322.They have no correlation.Conclusions:OPN may participate in the inflammatory response in acute stage of VMC and might promote the collagen synthesis in recovery stage.
