Objects Using reverse transcription polymerase chain reaction (RT-PCR)to clone human leukocyte antigen(HLA)-Ⅰmolecular recognizing antigen gene B, and analyze the gene expression of HLA-B from patients with breast carcinoma (BC)in peripheral blood leukocyte cells(PBLs).To explore the relationship between the HLA-B mRNA expression levels and the differentiation and lymph node metastasis of breast carcinoma in order to find the useful marker for the clinical evaluation of lymphatic metastasis, differentiation and prognosis for patients with breast carcinoma.Methods The special primers of HLA-B were designed and selected, glucose3-phosphate dehydrogenase(G3PDH)as inner reference, amplificated HLA-B mRNA and G3PDH mRNA by RT-PCR. Thirty-five patients of BC, including no lymph node metastasis, lymph node metastasis and stageⅠ-Ⅱgroup,Ⅲ-Ⅳgroup and 20 normal cases,20 cases of benign breast tumor patients were included in this study. Collection the cases'anticoagulant blood and extraction the PBLs, the amplified fragments were examined by 1.5% agarose gem electrophoresis。The IOD of HLA-B mRNA from PBLs were analyzed by gel analysis software Gel pro 4.0.Results1. The expression of PBL HLA-B mRNA in BC was significantly lower than in the benign breast tumor group and the normal control group. (P<0.01) 2. There was no significant difference between the expression of PBL HLA-B mRNA in the normal control group and the benign breast tumor group.(P>0.05)3.Compared with the expression of PBL HLA-B mRNA in the no lymph node metastasis group,the lymph node metastasis group significantly lower. (P< 0.05)4.Between the expression of PBL HLA-B mRNA in the stageⅠ-Ⅱof BC patients and the stageⅢ-Ⅳ,there were declined significantly. (P<0.05)Conclution1. The expression of HLA-B mRNA from PBLs in patients with breast carcinomas, especially for those with the lymph node metastasis or in the stageⅢ-Ⅳ, were lower, leading to default of antigen presentation, and hinting the relationship between HLA condition of host and metastasis and differentiation of tumors. In a word, the decline of HLA-B mRNA expression levels is possible an important factor for leading evasion of breat carcinoma from immune surveillance.2. It may reflect the degree of Progression and metastasis of lymph node in BC by detecting the expression levels of HLA-B mRNA,which may provide some instruction to early diagnosis,immune therapy and Postoperative follow-up of the breast cancer.
