Objective:This paper aims at researching on the changes of the Oxidative Stress, anti-oxidation defense and HOMS-IR of the IGR and T2DM patents, which brought by the synthetic intervention to them. From which we could provide theoretical support for the clinical control of the T2DM and reducing the diabetes incidence from IGR people. Methods:First, divide all the participants into three groups, namely NGT, IGR and T2DM groups, according the results of Oral Glucose Tolerance Test. Measure the total assay of MDA, GSH and SOD, in order to assess the Oxidative Stress, Anti-Oxidation defense of the participants, in which we use the stable model to assess the insulin resistance at the same time. Second, divide the willing participants of IGR and T2DM into two random groups as the control group and intervention group. Participants in the intervention group will take part in a one-year synthetic intervention, including three month comprehensive strengthening interference with food, physical exercise and health education in the very beginning, whereas there is no intervention to the control group. We will measure the physical situation, blood glucose, blood lipid and other relating characteristics of all the participants in two groups at the beginning and the end of the year. Then we will analyze the changes of all the results and oxidative stress level.Results:(1)In baseline case, the GSH and SOD of the T2DM group are obviously lower than those of the NGT and IGR groups. And MDA assay of the T2DM group is obviously higher than that of the NGT group. According to the analysis, the differences has the statistical significance (p<0.01, p<0.05). The HOMA-IR increases from NGT to IGR and to T2DM, which also has the statistical significance (p<0.01, p<0.05) based on the analysis. According to the analysis of the plasma insulin level and the overall situation of oxidative stress of the participants, results show that the GSH is negatively related with FBG,2hOGTT, HbAlC, HOMA-IR; and positively related with HDL-C. SOD is negatively related with FBG,2hOGTT, HbA1C.HOMA-IR is positively related with BMI, WC, FBG,2hOGTT, HbA1C and negative related with HDL-C. (2)At the end of the one-year intervention with the T2DM intervention group, the assay of GSH and MDA has increased and decreased respectively, which have statistical significance (p<0.01), and motility of SOD is lower than the baseline, whereas HOMA-IR increased and is higher than the baseline. The assay of GSH in IGR group kept on increasing and had great difference with the base line (p<0.01). The assay of MDA kept on decreasing and had great difference with the baseline (p<0.01). SOD activity decreased greatly (p<0.01) after three months'shape increase. HOMA-IR lowered in the third month and went up to the baseline at the end of the year. GSH and SOD of IGR group are obviously higher than those of the control group, which has the statistical significance (p<0.01).Conclusion:first, Oxidative stress may play an important role in the beginning and development of T2DM. Second, synthetic intervention can improve the oxidative stress and insulin resistance, thus can play an important role in the control of T2DM and prevention of IGR and provide the theoretical support to the clinical practice of the synthetic intervention.
