Objectives:Curcumin, a component of turmeric (Curcuma longa), is one such agent that is safe, affordable, and efficacious. Curcumin can kill tumor cells,but not nomal cells.β1,3-N-acetylglucosaminyltransferase 8 is a new member of glycosyltransferases we gained by electric cloning. We considered it belongs to beta-3 glycosyltransferase family.This research is focus on the relation between curcumin andβ3Gn-T8.Methods:To study the variation ofβ3Gn-T8 and its related genes expression in U251 and SMMC-7721 cell lines after curcumin added to the culture medium.Use RT-PCR to detect(1)the mRNA expression of MMP2,GnT2,GnT8 in U251 cell line and SMMC-7721 cell line;(2)the expression discrepancy of GnT2 and GnT8 in U251 cell line and SMMC-7721 cell line after curcumin added to the culture medium.Results After curcumin added to the culture medium,the mRNA expressions of GnT2, GnT8, and MMP2 in U251 and SMMC-7721 cell line decreased,and mRNA expressions of MMP2 decreased strongly.Conclusion This study showed that the expression of GnT2 and GnT8 are different in the two cell line after curcumin added to the culture medium.This result suggested that the effect of curcumin maybe change the sugar chains of glycoprotein in cell surface.
