Protective Effect Of Traditional Chinese Medicine Hu-qi Extractum On Radiation-induced Salivary Glands In Wistar Rats

Objective:The aim of this study was to provide the evidence of hu-qi extractum on the research and development of radiation-resistant.Assessed stimulus intensity on golden Hamster' oral mucous membrane,and then analyed the protective effect on the saliva glands' hispathologic tissue and function against 60Co radiation-induced injuries in Wistar rats.Methods:(1)By grading and observing the histopathological tissue section,we assessed stimulus intensity on oral mucous membrane.Hamster were randomly divided into the high dose group,the low dose group and the control group(12 hamster in each group),male and female in equal. In the drug two intervention groups,put 0.3ml compound or excipient into both cheek pouches while in the control group,the same dosage of distilled water and administration of vehicle was used.Once for every hour,four times a day.One hour after the last time given drug,distilled water rush the cheek pouch,Evaluated the Oral mucous membrane irritation.Repeated the procedure daily for thirty consecutive days,and at the end of the thirtieth administration, observed the pouches one hour,twenty-four hours,forty-eight hours and seventy-tow hours after the irrigation respectively,evaluated and graded the irriative response by local edema and erythema,and histopathologic examination of mucous membrane from oral cheek was performed.(2)The saliva glands of male Wistar rats were locally 60Co7-irradiated in the head and neck region with a single dose of 15 Gy and analysed the composition of the saliva samples,morphological changes at 3,40 days after irradiation.Male Wistar rats were randomly divided into the sham-irradiated control group(SI),the high dose group(HDG),the middle dose group(MDG),the low dose group(LDG) and the single dose irradiated group(SD)(20 Wistar rats in each group).All intervention rats were intragastric administrated with hu-qi extract,while the control group and model group were administrated with saline.Two phases of glands reaction to irradiation have been noted.Wistar rats were anesthetized,pilocarpine were injected intraperitoneally to stimulate saliva flow.Saliva was collected after pilocarpine injection.Assuming the specific gravity of saliva to be 1.0g/cm3 and saliva flow rate was recorded as flow per minute.All samples were stored frozen (-20℃)for further tests.The concentration of sodium,potassium,amylase were measured by atomic absorption spectrophotometry.SIgA were analysed by ELISA techniques.After sacrifice, the bilateral parotid glands and the submandibular glands were removed immediately and weighed.The morphology was assessed after H&E staining. Results:(1)Localized epitheliosis of the cheek mucosa had been found in single case from the high dosage group and control group,so it was assumed to be accidental and irrelevant to irritation. Since no pathological changes had been found in most cases,it was estimated that the extracts had no irritation on golden hamster's cheek mucosa.(2)The weight of the rats in single dose iiradiated group,low dose group,middle dose group decreased comparing with that of the sham-irradiated control group on the fortieth day,There was no significant difference between the high dose group and the control group.The SIgA decreased after irradiation (p<0.05),while no significant difference in those of the high dosage group which were close to the sham-irradiated control group.The amylase concentration of saliva of experimental group was significantly higher than that of the control group on the third day (p< 0.05).But there was no significant difference on the fortieth day between the three dosage groups and the single dose irradiated group.After irradiation,the cellular plasm of acinus of saliva glands showed the progressing changes of swelling,vacuolation and necrosis.The acinar showed atrophy and less cells compared with control at late time.Conclusions:Our results suggest that compound hu-qi extract which had no irritation on golden hamster's cheek mucosa and provided the safety for clinical application improve the general conditions and protect the structure and function of salivary glands in wistar rats undergoing radiation.
