The Effects Of "BuShen HuoXue" Acupuncture On The Expression Of Flotillin-1 And NEP In Hippocampal CA1 Region Of The SAMP8 Mice Mode

Objective:To investigate the effects of "BuShen HuoXue" acupuncture on the learning and memory abilities of the elderly aging SAMP8 mice mode and the expression of Flotillin-1 and NEP in its hippocampal CA1 region.Methods:20 6-month-old SAMP8 mice were randomized into 2 groups:SAMP8 control group and acupuncture group, with 10 mice in each group. Another 10 6-month-old SAMR1 mice were assigned to SAMR1 control group. The acupuncture group was treated by acupuncture at Baihui acupoin,Geshu acupoint and bilateral acupoint of Shenshu and Xuehai once per day for 4 weeks. At the end of the course of treatment, the learning and memory abilities were tested and the expression of Flotillin-1 and NEP in hippocampal CA1 region were detected by Immunohistochemistry.Results:1. The ability of learning and memory observation:The SAMP8 control group had significantly decreased in ability of learning and memory than the SAMR1 control group (the longer escape latency,the more times in platform quadrant);The acupuncture group had significantly increased in ability of learning and memory than the SAMP8 control group (the Shorter escape latency,the less times in platform quadrant).2. The investigation of immunohistochemistry:The SAMP8 control group had significantly increased in the level of Flotillin-1 expression(the higher IOD value) but significantly decreased in the level of NEP expression(the lower IOD value) than the SAMR1 control group; The acupuncture group had significantly decreased in the level of Flotillin-1 expression(the lower IOD value) but significantly increased in the level of NEP expression(the higher IOD value) than the SAMP8 control group.Conclusion:1. SAMP8 is reliable as a animal model of Alzheimer's.2."BuShen HuoXue" acupuncture can improve the learning and memory of the Senescence Accelerated Mouse Prone8. 3."BuShen HuoXue" acupuncture can prevent Alzheimer's disease by decreasing the Flotillin-1 leveland increasing the NEP level in hippocampal CA1 region, those partly may be therapeutic mechanism on Alzheimer disease of "BuShen HuoXue" acupuncture.
