Objective:To observe the effect of preventive anti-aging acupuncture therapy on animal behavior and hippocampus neuron shape of senescence accelerated mouse (SAM) P8, and explore the prevention of acupuncture to Alzheimer's disease.Methods:30 mice of seven-month old SAMP8 were selected and randomly divided into SAMP8 control group,acupuncture group and fixed group,10 age-matched SAMR1 mice were enrolled as control group. The acupuncture group was given the preventive acupuncture therapy once every other day for 12 weeks; the fixed group was given the same fixed action as the acupuncture group. Morris water maze tests were used to observe spatial reference memory ability of each group before starting of therapy and after each period, the effects of preventive acupuncture therapy on cognitive function in SAMP8 was analyzed. After behavior study, all of mice were perfused with 4% paraformaldehyde by aorta abdominalis. Paraffin sections were stained by Nissl technique to observe the change of hippocampus neuron shape in each group. Moreover, we used Image-pro Plus 6.0 to calculate the average grey of pyramidal cell layers in hippocampus CA1 field and analyze the effect of preventive acupuncture therapy on the form and function of hippocampus neuron in SAMP8 mice.Results:1 After three therapy period, compared to R1 control group, SAMP8 mice showed significant deficits on cognitive function in place navigation trial and spatial probe test (P<0.05). Acupuncture group showed a significant improvement in learning and memory when compared to P8 model group and fixed group(P<0.05), with no significant difference when compared to SAMR1 control group(P>0.05).2 R1 control group:the shape of neuron was regular, the cell layers had tight order, and the number of tigroid body was rich; P8 model group:the shape of some neurons was irregular, the cell layers were disorder, and the number of tigroid body decreased; The neuron shape of acupuncture group was in good condition, better than that for P8 model group. Compared to R1 control group, the average grey of pyramidal cell layers in hippocampus CA1 field of P8 model group decreased significantly(P<0.01);Acupuncture group showed a significant improvement in the average grey of pyramidal cell layers in hippocampus CA1 field, when compared to P8 model group(P<0.05), with no significant difference when compared to SAMR1 control group(P>0.05).Conclusion:1 Ten-month old SAMP8 mice were starting to represent obvious impairment on cognitive function. After preventive acupuncture therapy, ten-month old SAMP8 mice were mostly normal. It pointed out that preventive acupuncture therapy could have delayed action on the senile accelerated process in SAMP8 mice.2 The number of tigroid body in hippocampus neuron and the function of compounding protein in SAMP8 mice were decreased. After preventive acupuncture therapy, the hippocampus neuron of SAMP8 mice was mostly normal. It pointed out that preventive acupuncture therapy could improve the form and function of hippocampus neuron, decrease or delay the loss of neurons.
