The Research On Identification And Polysaccharide Extracting Optimization Process Of Radix Hedysari

Radix Hedysari is authentic Chinese herb medicine of Gansu. It recorded early in Shennong's Classic of Materia Medica, and it has a long application history. From the 1950s, scholars both at home and abroad studied to Radix Hedysari, and understanded it include polysaccharide, amino acids, flavonoids, alkaloids. The main active component Hedysarum polysaccharide (HPS) of Radix Hedysari could improve superoxide dismutase(SOD), thus enhance immunity, anti-aging, strengthens lymphatic factor activation killer cells (LAK) to tumor cells to kill and various biological effects.Now there were more researches on efficacy and pharmacology of Radix Hedysari, but many molecular biology mechanism of pharmacological effects were not clear, at the same time the extracting methods were not comprehensive. Based on single factor experiment and orthogonal orthogonal experiment to optimize the extraction process of Radix Hedysari polysaccharide. It provides the basis for the dustrialized production and the development of new dosage form.The paper authenticated appearance, microscopic structure of powder and slice. Preliminary experiments shows Radix Hedysari contains amino acid (or protein), saponins, organic acids, phenol, flavonoids, alkaloids, sterols (or terpene). We have gotten the optimum technics parameters that extracting polysaccharides from Radix Hedysari in the water-extracting experimeng is 80℃, sample:water=1:12, lhour, two times, ethanol precipitation concentration 70%. In this condition, the content and yield of polysaccharides is 64.23%,3.92%.At last, we have used cellulose enzyme extracting polysaccharides. Zymolysis concentration, zymolysis temperature, zymolysis time have been selected to be three key factors in direct cross experiments. The optimum conditions of cellulose enzyme extracting is dosage of enzymes 0.3%(quality score),40℃,110 minutes.At the same time, the same materials were adding to acidity water alkaline solution followed the optimum technics parameters, the result is the yield in both of the method were less than water-extraction. Then we compared with the content of polysaccharides in three plant such as planting herbs,wild herbs and astragalus membranaceus, the most content of three is wild Radix Hedysari.In the purification process, we main consideration defatted conditions. Defatted solvent(95%ethanol, methanol, petroleum ether), defatted time(60,90120min), and defatted time(60℃,70℃,80℃) have been selected to be three key factors, With crude polysaccharide and ratio of evaluation indexes.The optimum conditions of defatted conditions is petroleum ether,70℃,60 minutes.
