Objectives To investigate the association between -115A(rs12221497) and -6A(rs11039155) point mutation of liver X receptorαand the risk of coronary heart disease(CHD) in Han Chinese.Methods The -115A and -6A polymorphism in the liver X receptorαgene were detected by using single-labeled probe technique. Plasma cholesterol, triacyglycerol, HDL- cholesterol, LDL- cholesterol, fasting blood glucose(FBG) and body mass index(BMI) were determined 243 patients with CHD and 256 normal controls.Results (1) LXR-115A(rs12221497) have three different genetypes : genetypes of GG homozygote, AA homozygote, and GA heterozygote. LXR-6A(rs11039155) have only one genetype : genetypes of GG homozygote.(2) There was significant difference in frequencies of allele and genotype in -115A polymorphism between CHD and control groups respectively. The frequencies of the GG+AA genotypes and A allele were significantly (P<0.05 and P<0.01) higher in CHD patients than normal controls. Compared with GG genetype, the relative risk for CHD in people with GA genetype and AA genetype was 1.732(P<0.05).The -115A allele frequency was higher in CHD group than in cotrol group, the odds ratio (OR) (95% CI) of CHD is 1.81(95%CI:1.15~2.85), P<0.01 (3) Asscociation of -115A polymorphism with plasma lipid and FBG: There was no significiant associations between the -115A polymorphism and total cholesterol(TC), total triacyglycerol (TG), HDL- cholesterol, LDL- cholesterol, fasting blood glucose(FBG), body mass index(BMI). (4) In CHD patients, There was no significiant associations between the -115A polymorphism and total cholesterol(TC), total triacyglycerol (TG), HDL- cholesterol, LDL- cholesterol, fasting blood glucose(FBG), body mass index(BMI), constituent ratio of hypertension, diabetes mellitus,smoking history,gender, vascular lesion. Coronary artery angiography was done for all the CHD, According to the severity of coronary stenosis, CADi coronary system was derived. The scores of CHD with GA+AA and AA genotype(44.17±17.66) were same with RR genotype(44.21±18.26). (5) Binary logistic regression analysis showed that polymorphism of -115A was an independent risk factor for CHD. People with GA or AA genetypes take higner risk of suffering from CHD than people with GG genetype, the odds ratio (OR) (95% CI) is 1.83(95%CI:1.02~3.31), P<0.05.Conclusion The polymorphism of -115A of liver X receptorαgene could be associated with risk of CHD in Han Chinese.
