Objective To study the relationship between myeloperoxidase (MPO) and NAD(P) H:quinone oxidoreductase (NQOl) genetic polymorphisms and the susceptibility of the acute leukemia in Gansu population.Methods A 1:1 paired case-control study of 100 patients of acute leukemia and 100 cancer-free hospital subjects as a control group was conducted to detect myeloperoxidase (MPO) and NAD(P) H:quinone oxidoreductase (NQO1) polymorphisms by PCR-RFLP and PCR-LDR techniques.Results The A allele genotype and GA/AA genotype of MPO gene occurred more frequently in control group (28% and 54%) than those in AL group (19% and 31%), however, the T allele genotype and TC/TT genotype of NQO1 gene occurred more frequently in AL group (46.5% and 68%) than those in control group (28% and 43%). Further stratified analysis, the GA/AA genotype of MPO gene occurred more frequently in control group (54%) than those in AML group (28.2%). The controls had decreased 0.346-fold AML risk, compared with the Individual with GG genotype; the TC/TT genotype of NQO1 gene occurred more frequently in AML group (63%) than those in control group (43%). The people with TC/TT genetype had increased 1.67-fold AML risk, compared with the Individual with CC genotype. Combined genotype analysis showed that individuals carrying the GG genotype of MPO gene and the TT and TC genetype of NQO1 increase a 2.57-fold risk of AML, compared with the Individual with GA and AA genotype of MPO gene and CC genetype of NQO1 gene.Conclusion MPO and NQO1 genetic polymorphism was associated with susceptibility of acute myeloid leukemia in Gansu population. notypes may be used as a stratification marker to predict high-risk individuals for AML, the risk of AML was decreased in the persons carrying GA or GG genetype of MPO gene, however, the risk of AML was increased in the persons carrying TC or TT genetype of NQO1 gene. The individuals carrying the GG genotype of MPO gene and the TT and TC genetype of NQO1 further increase the risk of AML.
