Background: The synuclein family is made up of three disinct genes,alpha-synuclein is one of them which was initially discovered in Torpedo california,by expression cloning, using antibodies against cholinergic synaptic vesicles. It was named as such as it was localized to presynaptic nerve terminals and atrifactually to nuclei. Recent research about alpha-synuclein expression in alcohol addition has implicated that, alpha-synuclein proposed to serve as a negative regulator of dopaminergic neurotransmission. As we al known dopaminergic neurotransmission is the key of drug or alcohol use. The aim of our study was to observe the changes in alpha-synuclein expression after Acute Alcoholism or Chronic Alcoholism in rats, discussion mechanisms of alcoholism in Central nervous system.Methods: Male SD rats were divided into Acute Alcoholism and Chronic Alcoholism by random. The Chronic Alcoholism rats were given constant an 15% alcohol 4ml once a day for 30 days, and were decapitation 2h after the last feed. The Acute Alcoholism rats were given 50% alcohol, the amount of alcohol depends on rats'weight. According to the aim of our study, the rats were give 20g/kg alcohol,and were decapitation 1h,3h,6h,12h after the Acute Alcoholism model sucessfully prepared. Immunohistochemical staining were used to measure alpha-synuclein protein levels in the brain.Results:â‘ The HE staining shows there is cerebral edema, Subarachnoid hemorrhage, the number of nerve cells reduced in rats brain.â‘¡The alpha-synclein expression level were elevated in cerebral cortex and brainstem of rat in Chronic Alcoholism.â‘¢The alpha-synclein expression level were elevated in cerebral cortex of rat in Acute Alcoholism also.Conclusions:â‘ The HE staining shows that rats'brain were damaged by Chronic Alcoholism and Acute Alcoholism.â‘¡The immunohistochemical staining shows that the alpha-synclein expression level were elevated in cerebral cortex and brainstem of rat in Chronic Alcoholism.â‘¢The immunohistochemical staning indicates that the alpha-synclein expression level were elevated in cerebral cortex of rat in Acute Alcoholism also.
