| Part1 Effects of Jiao-tai-wan and its single constituent herbs on Treating Type 2 Diabetes Mellitus in RatsObjective: To study the effects of Jiao-tai-wan and its single constituent herbson treating type 2 diabetes mellitus in rats.Methods: The model of type 2 diabetes mellitus in rats was established by the injection of streptozotocin from tail vein and feeding with high fat and high caloric diet. Diabetic rats were randomly divided into model group, Jiao-tai-wan group, Rhizoma Coptidis group, Cinnamon group and metformin group. Rats in different treatment groups were given by corresponding therapy from gastric tube. Meanwhile normal control group was established. Body weight, oral glucose tolerance test (OGTT), blood lipids level including total cholesterol (TC), triglyceride (TG), high density lipoprotein-cholesterol (HDL-C) and low density lipoprotein-cholesterol (LDL-C), plasma levels of free fatty acid (FFA) and adiponectin were determined at the end of the eighth week of experiment.Results: Body weight, fasting plasma glucose (FPG), postpradial plasma glucose at one hour (PG-1h), postpradial blood glucose at two hour (PG-2h), plasma levels of TC, TG, LDL-C and FFA were increased in rats of model group compared with those in normal control group. Plasma levels of HDL-C and adiponectin were also decreased in model group (P<0.01). Compared with model group results of OGTT and blood lipid levels were improved while levels of HDL-C and adiponectin were increased in rats of different treatment groups (P<0.05 or P<0.01). Compared with metformin group blood level of HDL-C elevated significantly in rats of Jiao-tai-wan group (P<0.05). Compared with Rhizoma Coptidis group and Cinnamon group plasma levels of HDL-C elevated significantly in rats of Jiao-tai-wan group (P<0.05).Conclusion:It is suggested that Jiao-tai-wan and its single constituent herbs are effective on treating type 2 diabetes mellitus in rats. The effect of Jiao-tai-wan is better than that of Rhizoma Coptidis and Cinnamon.Part 2 Effects of Jiao-tai-wan and its single constituent herbs on ectopic fat accumulation in rats with type 2 diabetes mellitusObjective: To study the effects of Jiao-tai-wan and its single constituent herbs on ectopic fat accumulation in rats with type 2 diabetes mellitus.Methods: The model of type 2 diabetes mellitus in rats was established by the injection of streptozotocin from tail vein and feeding with high fat and high caloric diet. Diabetic rats were randomly divided into model group, Jiao-tai-wan group, Rhizoma Coptidis group, Cinnamon group and metformin group. Rats in different treatment groups were given by corresponding therapy from gastric tube. Meanwhile normal control group was established. Body weight, liver index, levels of fasting plasma glucose, fasting serum insulin and insulin resistance index (HOMA-IR), plasma activities of liver associated enzymes, triglyceride (TG) contents in hepatic, cardiac and muscular tissues, and their pathological changes were determined.Results: Body weight, liver index, activities of liver associated enzymes, HOMA-IR, TG contents in hepatic, cardiac and muscular tissues were all increased in rats of model group compared with those in normal control group(P<0.05 or P<0.01). Compared with model group HOMA-IR, plasma activities of liver associated enzymes and ectopic fat accumulation were improved in rats of different treatment groups (P<0.05 or P<0.01). Compared with metformin group contents of hepatic and muscular TG were obviously decreased in rats of Jiao-tai-wan group (P<0.01), muscular TG content was obviously decreased in rats of Rhizoma Coptidis group (P<0.05). Compared with Rhizoma Coptidis group and Cinnamon group plasma levels ofγ-GT decreased significantly in rats of Jiao-tai-wan group (P<0.01).Conclusion: It is suggested that the disturbances of glucose and lipid metabolism and abnormality of liver associated enzymes in type 2 diabetic rats could be improved by Jiao-tai-wan and its single constituent herbs. The therapeutic mechanism might be related to improving insulin resistance and reducing ectopic fat accumulation in non-adipose tissues. |
