Partâ… To Establish The Rat Model of Polycystic Ovarian SyndromeObjective: To establish the rat model of polycystic ovarian syndrome by giving intragastric administration of Letrozole.Methods: Twenty female SD rats were randomly divided into control group and model group. Model group was given intragastric administration of letrozole (0.1 mg/kg/d), while control group was given intragastric administration of 1% CMC once daily. The changes of estrous cycle were observed by means of vaginal smear. The rats were killed at 21 day later, weighed the ovary and observed the macroscopic shape, observed the microscopic ovarian follicle pathology alteration, and calculated the vesicular ovarian rate.The serum levels of hormones and FINS were detected by RIA. Results: Compared to control group, the rats in model group lost steady estrogen cycle and retained in interphase. The weight of the rats and two ovaries increased, and macroscopic examination showed thickened albuginea ovarii. Two ovaries showed the number of corpora lutea and the layer of granulosa cells decreased significantly, while the theca cells increased (P<0.01). The levels of testosterone(T), luternizing hormone(LH), fasting insulin(FINS) in model group increased significantly(P<0.05), the level of progesterone(P) decreased significantly in model group(P<0.05).The level of follicle stimulating hormone(FSH) decreased in model group, but had no statistical significance(P>0.05).Conclusion: letrozole intragastric administration can build an ideal rat model with polycystic ovary syndrome. Partâ…¡study on therapeutic effect and mechanism of acupuncture in treating PCOS ratsObjective: To observe the therapeutic effect of acupuncture on rat with polycystic ovarian syndrome and explore the mechanism of action.Methods: Forty female SD rats were randomly divided into two groups, 10 in control group and 30 in model group. Model group was given intragastric administration of letrozole (0.1 mg/kg/d) for 21 days, while control group was given intragastric administration of 1% CMC once daily for 21 days. After the PCOS model was established, the rats in model group continued to be given intragastric administration of letrozole(0.05 mg.kg-.d- ), and randomly divided into three group, model group, acupuncture group and western medicine group,10 rats in each group. The rats in acupuncture group were given acupuncture treatment once daily, LiuZhen for 15min. The rats in western medicine group were administered clomiphene citrate (CC) at the dose of 4.5 mg/kg/d. All rats were killed at 14 days later, weighed the ovarian, observed ovarian follicle pathology alteration. The serum levels of hormones and FINS were examined by RIA. The expression of Fas, FasL and PCNA in ovarian antral follicle was observed by immunohistochemistry and the apoptosis was by TUNEL methods.Results:â‘ Compared to control group, the weight of rats in model group and two ovaries increased, albuginea ovariisteady thickness increased. The ovary showed multiple follicular alteration, the number of corpora lutea and the layer of granulosa cells decreased significantly, while the theca cells increased. In acupuncture group, albuginea ovariisteady thickness altered little, and the number of corpora lutea and the layer of granulosa cells increased significantly, the theca cells and the folliculi ovarici vesiculosi decreased, compared with model group. In western medicine group, the folliculi ovarici vesiculosi decreased, the layer of granulosa cells increased, various kinds of ovarian follicles increased, the number of corpora lutea increased, and unruptured luteinizing cysts existed in some ovaries.â‘¡Compare with control group, the serum level of T, LH and FINS in model group increased significantly (P<0.01), while the level of FSH decreased obviously (P<0.01). Compared with model group, the level of T and FINS in acupuncture group and western medicine group decreased obviously (P<0.01), the level of FSH increased significantly (P<0.05), the level of LH decreased, but had no statistical significance.â‘¢Compare with control group, the expression levels of Fas and FasL increased significantly (P<0.01), while the expression level of PCNA decreased significantly (P<0.01). The expression levels of Fas and FasL in acupuncture group and western medicine group decreased significantly compare to model group (P<0.01), but were higher than that in control group (P<0.05). The expression level of PCNA in acupuncture group and western medicine group increased significantly (P<0.01), but was lower than that in control group (P<0.01). Compare with western medicine group, the expression level of FasL in acupuncture group decreased (P<0.05), others index had no statistical significance. The TUNEL results showed that ovarian follicle rate in model group was 45.16%,that in control group was 28.57% (P<0.01).Compared with model group, the ovarian follicle rate in acupuncture group and western medicine group decreased significantly (P<0.05), but there was no statistical significance between acupuncture group and western medicine group (P>0.05).Conclusion: Both acupuncture and clomiphene could adjust disorder endocrine level of PCOS rats, promote the growth of ovary. The therapeutic mechanism may correlate to regulating the expression of Fas, FasL and PCNA in ovary.
