Objective:To establish a standard tissue bank of genitourinary tumor.Methods:5-ml peripheral venous blood was collected preoperatively, anticoagulated byEDTA,and stored at -80℃.100-ml preoperative morning urine was collected from patients ofbladder cancer,then after concentration and centrifugation,concentrated urine and sedimentneeded to be preserved classifiedly.After resection of specimens,carcinoma tissues and theirsurrounding tissues were collected to be frozen, dealt with RNAlater, or fixed by formalin andstored at appropriate temperature and containers in accordance with standard operatingprocedures.Then we recorded the clinical data of patients, established scientifically the rigorousquality assurance and quality control system,and managed the specimen bankinformationizedly.Results:From March 2009 to April 2011,by full use of the good cooperation between ourlaboratory with urology department of seventeen large hospitals fromShenzhen,Changsha,Guangzhou,Hefei,Zhengzhou,Haikou and Shantou,more than 980 bloodsamples,1,467 tissue samples and 180 urine samples were collected from patients of urinary andreproductive system tumors,part of which had been used by more than 50 times for wholegenome sequencing,RT-PCR,immunohistochemistry, Western blot and methylation.Involvingsubjects included 863 projects,the National Natural Science Foundation,the Provincial NaturalScience Foundation,and postdoctoral,doctoral and master's graduate program.Feedback fromspecimen users and quality control results showed that specimens of the tissue bank couldsatisfy the need of molecular biology experiments.Conclusions:My laboratory had established the first standardized tissue bank of urogenitaltumors in Shenzhen, which provided a good service platform for research on urogenital tumors.
