Cortex Ilicis Rotundae is the drying tree bark or root bark of Holly Family, and its taste is bitter, and its natre is cold, and itan clear away the heat-evil and expell superficial evils and eliminate swell and alleviate pain, damp elimination, dispell wind-evil and neutralize poison, its synonym is Aquilaria sinensis (Lour.)Gilg, it record in the book of culling traditional Chinese medicine in the south mountain at first,which it is a civilian commonly used herb medicine in the south of china.The quality standard of Holly Bark.has not been reported. In order to develop a new use of Cortex Ilicis Rotundae, the quality standard and fingerprint of Cortex Ilicis Rotundae has been studied in this paper.The main contents were divided into two sections as follows:1. The quality standard of Cortex Ilicis Rotundae.This experiment research into the quality standard control of Cortex Ilicis Rotundae. andestablish the methods of quality estimate of Cortex Ilicis Rotundae.The routine methods was used of pharmacognosy such as freehand section and surface sliceto observe the microscopic characters of the leaves and stems of Cortex Ilicis Rotundae.and thin layer chromatography (TLC) to identify it. The results showed the plant is Ilex rotunda Thunb.According to technical requirements in the appendix in China Pharmacopoeia (Ⅰ, 2005 ED).The water content of 7.5~11.0%, total ash content of3.7~6.9% and alcohol extract content of 28.6~38.8% was obtained.We also use HPLC to detect the content of syringin and pedunculoside. The average content of syringin was 2.1%. The average content of pedunculoside was 9.8%.2. The fingerprint of Cortex Ilicis Rotundae. The fingerprint method based on high-performance liquid chromatography for the quality control of Cortex Ilicis Rotundae were firstly described and optimized. HPLC system was used to obtain the fingerprint spectrum of Cortex Ilicis Rotundae. Thechromatography condition was Dikma C18column(200×4.6mm,5μm),mobile phase was Methanol-0.1% phosphoric acid,and flow speed was 1.0 mL·min-1, detection wavelength was 210 nm, column temperature was 35℃.14peaks ascertained as the common peaks of the fingerprint form its unique fingerprint.In this experiment, the common fingerprint has been established and the similitude degree of 10 batches Cortex Ilicis Rotundae. has also been obtained (0.925~0.984).The facts mentioned aboved illuminate that these experiments successfully build thequality standard of Cortex Ilicis Rotundae and the fingerprint of Cortex Ilicis Rotundae by HPLC.The method is accurate, reliable, good reproducibility, strong characteristic and can be used for the standard of genera judgement and the quality estimation about the Cortex Ilicis Rotundae.
