Periodontal disease is one of the most common dental diseases, the incidence of which in our country is more than 70%. Periodontal disease often results in loss of teeth, thus affecting the patient's chewing function and reducing their quality of life. The current conventional treatment of periodontal disease is mainly dependent on mechanical operation, but still lack of ideal treatments. Alveolar bone resorption and formation of periodontal pocket are considered to be its main symptoms. Alveolar bone resorption is the major reason of tooth mobility and loss. Recent studies of the mechanism in bone resorption have found that OPG/RANKL/RANK axis is the key factor in the regulation of osteoclast differentiation and bone resorption. The system plays an important role in the pathogenesis of periodontitis. Researching the significance of OPG/RANKL/RANK system in the alveolar re-absorption can provide a new direction in the treatment of periodontal disease.Objective:Establish the model of alveolar bone resorption in experimental rat to research the law in bone re-absorption while observing the expression of OPG. RANKL. RANK. To understand the significance of OPG/RANKL/RANK system in the alveolar re-absorption.Methods:Alveolar resorption model was established by injection lipopolysaccharide of E. coli O111:B4 to the mesial gingiva of the first molar on the left mandible of SD rat. Research the law in the alveolar re-absorption. Detect the expression of OPG,RANK, RANKL by S-P Immunohistochemical.Result:There were obvious alveolar bone resorption and TRAP stained osteoclasts in the samples of E-LPS induced SD rat model, and the resorption effect was strong in the first 20 days. The OPG level in periodontal tissues was decreased compared with control group; RANKL and RANK were significantly noticed in the osteoclasts. The effect of alveolar resorption was reduced after 20 days.Conclusion:Expression of OPG. RANKL, RANK changed similar with alveolar reconstruction process and the number of osteoclasts. OPG/RANKL/RANK system plays an important role in bone resorption mechanisms. Objective:Explore the effect of periodontal-endodontic combined treatment on severe periodontitis patients.Methods:Three teeth with severe periodontitis were treated with perfect root canal treatment and periodontal treatment.Results:After 6 months-1 year follow-up observation, the function of the three teeth is recovered well.Conclusion:Periodontal-endodontic combined treatment has a positive effect on periodontal tissue regeneration in severe periodontitis...
