Objective To study the relationship between expression of stathmin and differentiation and metastasis of brain astrocytoma. To investigate the bioinformatic significance of stathmin as a biomarker of brain astrocytoma.Methods The expression of stathmin in specimens of 62 of brain astrocytoma, 20 of normal brain tissue come from autops or cerebral trauma specimens and were detected by immunohistochemistry. The relationship between stathmin expression and grading and the optimum and malignancy of astrocytoma. And transwell cave experiment were used to detected the effect of downregulation of stathmin caused by siRNA.Results: 1) The positive ratio of stathmin in brain astrocytoma and normal brain tissue were 77.4%(48/62), 30.0%(6/20), respectively. There were significance between them(P<0.05).2) The positive ratio of stathmin in astrocytoma I, II grade and astrocytoma III, IV grade were 46.2%(12/26), 100%(36/36), respectively. And there were significant difference in the expression of stathmin between theose groups(P<0.05).3) Transwell cave experiment were used to detected the effect of U251 cell line with downregulation of stathmin. And downregulation of stathmin caused a decreased of metastasis ability of U251 cells in vitro.Conclusion: 1) There were relationship between the expression of stathmin in astrocytoma and normal brain tissues. And the expression of stathmin may correlate to the development and progress of astrocytoma.2) There were relationship between the expression of stathmin and the differentiaion and astrocytoma I, II grade and astrocytoma III, IV grade, and could serve as a biomarker of differentiaion and grade in astrocytoma.3) There a direct proportion relationship between the downregulation of stathmin and decreased invisive ability of U251 cells in vitro, suggested that there were tightly relationship between stathmin and invasion of astrocytoma, and stathmin could serve as a biomarker of metastasis of astrocytoma.
