Naringin, hesperidin and neohesperidin are kind of important pharmaceutical bioactive ingredients of traditional Chinese medicines (TCM), used to be antioxidant, anti-hyperlipidemics, anti-atherosclerotics, and etc. Meantime, they are also acted as sweeteners, antioxidants, pigments, health care products in food industry. Naringin, hesperidin and neohesperidin are widely distributed in Rutaceae Citrus plant, such as Citrus Reticulatae Pericarpium, Citrus Reticulatae Pericarpium Viride, Aurantii Fructus, Aurantii Fructus Immaturus, Citri Grandis Exocarpium and Citri Sarcodactylis Fructus. In this dissertation, the author mainly studied on the extraction, separation, detection and distribution discipline of naringin, hesperidin and neohesperidin in some TCM, in which the goal want to provide a theoretical basis for effective extraction and quality control of TCM. This article is divided into four chapters, the main contents and results are summarized as follows:Chapter I, the physical / chemical properties, the main biological activity of naringin, hesperidin and neohesperid, and their distribution in natural plant were introduced. The authors reviewed the recent progress about the extraction, separation and determination of naringin, hesperidin and neohesperidin in Rutaceae Citrus plants, and then put forward the research goal of this dissertation.Chapter II, the extraction and separation of naringin and neohesperidin in Aurantii Fructus were studied by ultrasonic assisted extraction, Soxhlet extraction and reflux extraction, respectively. The influential factors on the extraction efficiency, such as solvent, time and temperature, were respectively investigated. The advantages and disadvantages of different extraction methods were also compared. The results demonstrated that the optimized conditions of naringin and neohesperidin in Aurantii Fructus by reflux are achieved below: extraction solvent 80% ethanol, extraction time 40min, extraction temperature 60℃. The optimized extraction solvent is 95% ethanol or methanol by Soxhlet extraction or by ultrasonic extraction, respectively. Comprehensive analysis concluded that microwave-assisted extraction or reflux extraction in water bath can improve the extraction yield, which is recommended for the preparations of production. Microwave-assisted extraction or ultrasonic-assisted extraction is preferred in drug analysis because of the more time-saving, convenient and quick.Chapter III, the extraction and separation of naringin in Aurantii Fructus were studied by microwave assisted extraction and HPLC as detecting method. The factors related to extraction, such as solvent, time, temperature and the power of microwave extractor were respectively investigated. The results showed that the optimized conditions by microwave-assisted extraction of naringin in Aurantii Fructus are achieved below: 80% ethanol as solvent, extraction time 10min, extraction temperature 40℃, microwave extraction device power 400W. Compared with reflux extraction in water bath, microwave-assisted extraction can short the extraction time and decrease the temperature.Chapter IV, the difference and dynamic variation discipline between the components of various TCM and raw materials were respectively investigated by comparing the contents of naringin, hesperidin and neohesperidin in various TCM and Rutaceae Citrus. The results indicated that the presented methods can be used for identifying the certified products or counterfeit of decoction pieces by detecting those components of TCM, which might meet quality control.
