Background and Objective:Acute leukemia is a clonal malignant disease of hematopoietic cells characterized by the excessive proliferation and inhibition of apoptosis. The mainly remission induction therapy of acute leukemia is cell killing, also induction of differentiation and apoptosis. but many patients can not be remission or relapse. In order to improve the remission and reduce the relapse rate of leukemia patients, increase the chemotherapy drug dose and drug combination as an iomport method, but with the chemotherapy drug dosage increases, its toxicity increased significantly. Matrine is an alkaloid extracted from Sophora flavescens. It has anti-inflammatory, immunosuppressive, antiarrhythmic, antiviral and resistance to parasitic effects, now its anti tumor effect is increasingly valued. Basic research confirmed that matrine could inhibit leukemia cell proliferation and induction leukemia cell apoptosis. but matrine injection combined with chemotherapy in acute leukemia study is very less, Therefore, we conducted a clinical and experimental research, to study its therapeutic effect in acute leukemia and its possible mechanism,provide new ideas for acute leukemiaMaterials and method:Select eighty cases untreated patients with acute myeloid leukemia, divided into two groups, each group of40patients.The experimental treatment group treat by DA program combined with matrine injection; the control treatment group only using the DA program. During chemotherapy monitor every patient blood analysis, liver and kidney function and bone marrow examination, and statistical differences. ELISA detect serum survivin protein levels; detect survivin expression of bone marrow mononuclear cells by Immunocytochemistry of those patients. ELISA detect survivin levels of the bone marrow mononuclear cell culture supernatants with different concentrations of matrineResults:The matrine chemotherapy can significantly improve the quality of life in patients,reduce chemotherapy-induced infection rates. Combined with chemotherapy can significantly reduce the serum survivin level. The positive expression of survivin protein in patients with acute myeloid leukemia bone marrow cells significantly increased The matrine can reduce acute leukemia bone marrow mononuclear cell culture supernatant of survivin protein levels which is negatively correlated with matrine.Conclusion:Matrine can inhibit leukemia cell proliferation and induce apoptosis in leukemic cells, in combination with chemotherapy drugs its can increase the chemotherapy drug efficacy, reduce side effects of chemotherapy, improve patients’quality of life. Matrine can reduce the bone marrow and serum of patients with acute leukemia survivin protein expression levels.
