0bjectivesTo investigate the effcts of procyanidin on rat myocardial inflammatory infiltration and TNF-alpha(tumor necrosis factor-α,TNF-α) expression.MethodsThe rat myocardial infarction model was established by left anterior descending coronary artery ligation. Survival rats were randomly divided into the myocardial infarction control group(MI n=9) and the procyanidin group(PC n=9,100mg/(kg.d)),and another sham group(sham n=10), Each group further divided into by 2weeks and 4weeks: MI2w and 4w,PC2w and 4w,sham 2w and 4w ; myocardial pathological structure in the infarcted and non-infarcted zone were observed at 2 and 4 weeks after MI with histopathological methods, The expression of TNF-αwas detcted by semi-quantitative RT-PCR, the expression of TNF-αwas detcted by immunohistochemistry and Western blotting. Hemodynamics is detcted by more conductive physiological instrument ( LVRW;LVEDP; +dp/dt; -dp/dt). ResultsAfter MI 2 and 4 weeks,(1)compared with sham group,major anomalies of myocardial pathological structure in MI and PC groups;compared with MI group, noticeable improvements of myocardial pathological structure in PC group.(2)compared with sham group, the gene and protein expression of TNF-αwas higher significantly in MI and PC groups (P<0.01).(3)compared with MI group , the gene and protein expression of TNF-αwas lower significantly in sham and PC groups(P<0.01).(4)MI Compared with sham group, hemodynamics is lower, +dp/dt -dp/dt is lower significantly, but LVEDP is higher significantly ,Hemodynamics and LVRW are better of PC group.Conclusions1.Procyanidin can attenuate the myocardial inflammatory infiltration and suppress the expression of TNF-α.2.Procyanidin can improve hemodynamics of rat myocardial infarction.
