ObjectiveTo evaluate the effects of procyanidin on cardiocyte apoptosis ofmyocardial ischemia-reperfusion injury in rats and its probable mechanism.MethodsFourty male SD rats were randomly assigned to four groups: shame group,ischemia and reperfusion group,low-dose group(procyanidin50mg·kg-1)and high-dose group(procyanidin100mg·kg-1).The rats were pretreated byintragastric administration for2weeks. All rats,except for those in shamegroup,received occlusion of left anterior descending artery (LAD) for30minutes followed by120minutes of reperfusion.Serum CK-MB were detectedafter experiment.The ROS fluorescent probe dihydroethidium was used for themeasurement of ROS.The expression of p53, caspase-9was determined bywestern blotting. The myocardial apoptotic index was measured by the methodof TUNEL.ResultsCK-MB: Ck-mb activity increased significantly in the I/R group Comparedwith the shame group. Compared with the I/R group, PC50and PC100reducedck-mb activity, while ck-mb activity in PC100group was lower than the PC50group.ROS: ROS activity increased significantly in the I/R group Compared with the shame group. Compared with the I/R group, PC50and PC100reduced ROSactivity, and ROS activity in PC100group was lower than the PC50group.P53and caspase-9: The exppression of P53and caspase-9was increasedin the I/R group compared to the shame group, and PC50and PC100decreased the expression of P53and caspase-9compared with the I/R group,and,PC100decreased the expression of P53and caspase-9compared with thePC50group.Apoptotic Index: AI increased significantly in the I/R group compared withthe shame group. Compared with the I/R group, PC50and PC100reduced AI,while AI in PC100group was lower than the PC50group.ConclusionProcyanidin can reduce reduced apoptosis index afterischemia-reperfusion. This beneficial effect is partially dependant onROS-p53-caspase-9signal transduction pathway....
