Objective: Chronic atrophic gastritis is frequent diseaes of digestive system which was classified as precancerous changes in 1978. The main theat method is to restrain gastric acid, protect stomach mucosa, restrain Hp, ect. On the base of feature and pathogenesis and change of life style of people, combining many years clinical experience professor li produce the theory of"Toxic cloud". Using the theory to theating CAG the result is predominance and"Toxic cloud"intrinsic is main type of CAG, with improving of living standard of people. It is not only a factor todamage body, but also a pathoproduce."Toxic cloud"damage body, in change of morphosis of cell, structure, ect. In pathematology, it is hyperplasy, overgrowth, analosis, ect. The pathogenesy of CAG is attack and protect factor error, in controlling of nervousness-secretion-immunifaction system. MOT can shrink gaster, intestine, infraesophagus of esophagus, ect. SS Generally restrain function of gastrointestinal tract. The pathogenesis of CAG is error of attack and protection factor which is controled by nervousness-secretion-immunity Lattice. The archi-contribution of MOT is encouraging motivation of stomach intestine and gastric emptying (GE). SS distribute in brain and GE, its effect is adverse. By observing change of gut hormone and stomach intestine Motivation which is effected by JieDuHuaZhuoHeWeiFang, this experiment approach mechanism of JieDuHuaZhuoHeWei Fang on CAG rats. In order to explore this effect and mechanism we carried out an experiment study on animals.Method: Feeding Wistar 66 rats one week adaptatively, the weight of rats is between 150-170g, then in accordance with random number table, 13 randomly selected as the normal group, and the remaining are model. The remaining rats in each group give 2% sodium salicylate and 60% ethanol mixed solution gavage, 20mmol/L sodium deoxycholate, combining with hunger satiety disorders, 90 days later the models were created successfully. In the process of these 16 rats dead (six in them are killed by obersoering the effect of model). There maining 40 were randomly divided into 4 groups: model group, Victoria Su-enzyme group, JieDuHuaZhuoHeWeiFang high-dose group (referred to as high-dose), JieDuHuaZhuoHeWeiFang middle-dose group (referred to as midlle-dose group), JieDuHuaZhuohewei Fang low-dose group (referred to as low-dose group), 8 each group. After the rats model of CAG are successful, treatment was given the next day: the normal control group and model group were given equal volumesaline, control group were fed with Victoria Suenzyme suspension which density is 0.1g/mL, 4mL/d, twice one day. High dose group fed with JieDuHuaZhuo HeweiFang decoction, which including 25g/kg/d, middle-dose group fed with JieDuHuaZhuoHeWeiFang decoction, which including 12.5/kg/d. low-dose group fed with JieDuHuaZhuoHeWei Fang decoction, which including 6.25g/kg/d. Each group administer for days. At the end of fasting times after administration, cut-water 18 hours, intragastric administration, after 40 minutes, diethyl ether anesthesia, carat abdomen, unloading stomach intestine, decapitated blood, each 6mL, to do the following indicators: somatostatin (SS), motilin (MOT), gastric emptying rate (GER), intestine facilitation rat (IFR), and collecting specimens of gastric determination to do the following indicators: Gross observation of gastric mucosa, gastric muc- osa pathological observation.Results:1 General condition observationIn modle group the fur of rats is bloom, mind dispirited, reaction slow, appetite is bad, sed is abnormal, weight decrease, ect. Every theating group all can improve weight, appetite, ect. The high-dose of JieDuHuaZhuoHeweiFang is prominent.2 Gastric general observationAt the end of the experiment we found that the model group the color of gastric mucosa is whitish or gray, plica reduced or disappeared, less mucus, some are accompanied by scattered bleeding points and ana brosis. After therapies by Victoria Su-enzyme, low-dose, Mid-dose and high-dose JieDuHuaZhuoHeWeiFang, gastric mucosal damage have different improvement, the most obvious is high-dose group. The normal group, the color is pink, plica regular.3 Gastric mucosa pathology observationIn modle group the the thickness of gastric mucosa is normal, the gastric mucosa intrinsic glands reduce and disorder, the volume is not same. Endothelial cell fall off, lots of lymph cells and orther inflammatory cell in matrix, ect. After therapy of Victoria Su-enzyme, low-dose herb, mid-dose herb and high-dose herb, gastric mucosa improve and the most obvious is high-dose group.4 Stomach intestine hormone4.1 The impact on somatostatin (MOT)In model group serum MOT is significantly lower than the normal group, the two have significant differences in comparison (P<0.05); JieDuHuaZhuo- HeWeiFang high-dose, middle-dose and low-dose group and Victoria Su-enzyme group can rise serum MOT content significantly, compa- ring to model group there is significant difference (P<0.05), it shows that Jie- DuHuaZhuoHeWeiFang and Victoria Su-enzyme group can control serum MOT content effectively; In the four treating groups serum MOT concen rise, comparing with the normal group there is significant differences (P<0.05). comparing low-dose group with Victoria Su-enzyme group, serum MOT cont- ent have significant difference (P<0.05); there is significant difference compa- ring high-dose with low-dose (P<0.05), and no sifnificant difference between middle-dose and high-dose (P>0.05).4.2 The impact on somatostatin (SS)In model group serum SS is significantly higher than the normal group, the two have significant difference in comparison (P<0.05); JieDuHua- ZhuoHeWeiFang high-dose, middle-dose and low-dose group and Victoria Su-enzyme group can cut down serum SS content significantly, comparing to model group there is significant difference (P<0.05), it shows that JieDuHuaZhuoHeWeiFang and Victoria Su-enzyme group can control serum SS content effectively; In the four treating groups serum SS concen loss, comparing with the normal group there is significant differences (P<0.05). Comparing low-dose group with Victoria Su-enzyme group, serum SS content is nosignificant difference (P>0.05); there is significant difference (P<0.05), comparing high-dose with low-dose and no sifnificant difference between middle-dose and high-dose.5 Stomach intestine motivtion5.1 The impact on gastric emptying rate (GER)In model group GER is significantly lower than the normal group, the two have significant difference in comparison (P<0.05); JieDuHuaZhuoHe- WeiFang high-dose, middle-dose and low-dose group and Victoria Su-enzyme group can rise GER significantly, comparing to model group there is significant difference (P<0.05), comparing with the normal group there is significant differences (P<0.05). Comparing low-dose group with Victoria Su-enzyme group, there is no significant difference (P>0.05); there is significant difference comparing high-dose with low-dose (P<0.05) and no significant difference between middle-dose and high-dose.5.2 The impact on intestine facilitation rat (IFR)In model group IFR is significantly lower than the normal group, the two have significant difference in comparison (P<0.05); JieDuHuaZhuoHeWei- Fang high-dose, middle-dose and low-dose group and Victoria Su-enzyme group can rise IFR significantly, comparing to model group there is significant difference (P<0.05), comparing with the normal group there is significant differences (P<0.05); comparing low-dose group with Victoria Su-enzyme group, there is no significant difference (P>0.05); there is significant difference (P<0.05), comparing high-dose with low-dose and no sifnificant difference between middle-dose and high-dose (P>0.05).Conclusion: This experiment proved that JieDuHuaZhuoHeweiFang have the following effects: 1 improve the morphology of gastric mucosa histopathology; 2 rise expression leve of MOT and degrade level of SS of CAG rats; 3 improve stomach intestine motivtion of CAG rats; 4 the theat mechinism of JieDuHuaZhuoHeweiFang is to control the level of stomach intestine motivtion and stomach intestine hormone.
