Objective: Colorectal carcinoma (CRC) is one of the most common tumors of gastrointestinal tract. Recurrence and metastasis in tumor are the significant factors that affect the prognosis in CRC Patients. In this study, we determined the expression of Ezrin, simultaneously microvessel density (MVD) of circulating tumor cell and the number of circulating tumor cell. And then analyzed the significance of unnormal expression of Ezrin, the microves- sel density and the number of free cells in the blood, to investigate the relation of these factors and liver metastasis in colorectal carcinoma.Methods: The experiment randomly collected the inferior mesenteric blood of 48 cases with CRC after surgical therapy and the peripheral venous blood of 15 normal cases as comparison, which collected on empty stomach in morning. The blood were determined the cell of CD55 and CK19 double positive as the maker to detect the quantity of free cancer cell of CRC by flow cytometry. Meanwhile, the expression of Ezrin and the microvessel density labelled by CD31 monoclonal antibody through immunohistochemistry staining. And to analyze the correlation of the quantity of circulating tumor cell of CRC, the expression of Ezrin and microvessel density with gender, age, the site of cancer, level of differentiation, TNM stages, lymph node metastasis, liver metastasis.Results: In the 48 cases with CRC, the positive expression of Ezrin was 39.58%, and was statistical significance compared with the normal mucous membrane by increasing 6.67%(P<0.05). The positive expression of Ezrin in the CRC with liver metastasis was 70.00%, which conspicuous increased compared with the non liver metastasis 31.58% (P<0.05). The positive expression of Ezrin in the CRC with lymph node metastasis was 50.00%, was conspicuous increased comparing with the non lymph node metastasis 18.75% (P<0.05). There were no statistical difference in the expression of Ezrin of different gender, age, the site of tumor and the differentiation of tumor (P>0.05). There was a statistical difference betweenⅡandⅣof TNM stages (P<0.05), but no statistical significance in other TNM stages (P>0.05).In the tissue of 48 cases with CRC, the result of MVD was increased notably(20.34±2.63), compared with the normal mucous membrane (8.8±2.47) (P<0.05). In the tissue of CRC with liver metastasis, the result of MVD was increased distinctly (23.43±2.34), compared with the non liver metastasis (18.74±2.63) (P<0.05). There were no significant difference about the results of MVD in different gender, age, the site of tumor, the tumor differentiation, TNM stages and lymph node metastasis(P>0.05).In the 48 cases with CRC, the double positive of CD55 and CK19 (3.52%) increased notably(P<0.05), compared with the healthy people(0.06%). The double positive of CD55 and CK19 of CRC with lymph node metastasis (4.83%) conspicuous increased(P<0.05), compared with the non lymph node metastasis (0.44%). And it was 5.28% with liver metastasis, which conspicuous increased comparing with the non liver metastasis 0.47% (P<0.05). There were no significant difference about the double positive of CD55 and CK19 in different gender, age, the site of tumor and the differentiation of tumor (P>0.05). There was a statistical significance betweenⅠandⅣof TNM stages (P<0.05), but no statistical significance in other TNM stages (P>0.05).The relation of the MVD and the double positive of CD55 and CK19 was positive correlation (r=0.63, P<0. 05). And There was aslo positive correlation between the expression of Ezrin and the detection rate of CD55 and CK19 cell (r=0.72, P<0. 05).Conclusion:1 The expression of Ezrin of the tissue of CRC increases remarkably, compared with the normal tissue. And even higher in the tssue accompanying with lymph node metastasis and liver metastasis.2 The expression of MVD of the tissue of CRC increases remarkably, and even higher in the tissue accompanying with liver metastasis. And the microvessel density takes an advantage for liver metastasis.3 The detection rate of CTC is higher than the contrast control and characterizes highing tendency accompanying with metastasis.4 Simultaneous determination the expression of Ezrin, MVD and CTC is helpful to evaluate the aggressive potentiality of CRC, which might be one of the biological marker for the aggressive potentiality of CRC.
