| Objective:We Observed the effect of Tian Bing Tiao Du Capsule to affect the amount of food intake, volume of Fluid intake, Hobby ratio, emotional behavior, Spatial memory,and substance p of the rats, study the Nature of Disharmony of Liver and Stomach,so as to reveal mechanism from the neural physiological chemistry point of view.Methods:We divide the 60 minimal disease Male wista rats provided by The Experimental animal centre of He Bei Meidical University in to six groups:the normal group(A),the irritation and stress control group(B),the Deanxit group(C),the chai hu shugan group(D), the traditional Chinese medicine (Tianbing Tiaodu Capsule) high-dose group,the traditional Chinese medicine (Tianbing Tiaodu Capsule) low-dose group.All the rats can eat and drink freely.The rats'food is provided by the Experimental animal center of He Bei Medical University.We measured the amount of food intake and volume of Fluid intake everyday on 9:00 am. According to the report,the model groups were maken by Clamping 1/3 distal end tail by ring clamp, 30 seconds each stimulus, interval 3 hours each stimulus, 3 times each rats one day. The rats have rest for half an hour after stimulus and raising for separate.The rats in the model groups can be easily burned up and will bite each other.Compared with the normal group ,the model ones have less food and Depressed everyday,with its fur dry and yellow. The stimulus continued for 7 days.After 7 days,we measure the rats'weight,the amount of food intake,and the emotional behavior to test whether the model is succeedly made.Using the one-way anova,we discovered that there is a sgnificant differences between the control group and normal group.It proves that the model is successfully made.After observation and stimulus for 7 days, the rats of normal group and control group were given 2ml distilled water through watering stomach once a day.The chai hu shu gan group were feed concentrated decoctio of chai hu shu gan 1.75g/2ml/100g.The Deanxit group were feed suspension of deanxit 5mg/2ml/100g.The low-dose group were feed suspension of Tian Bing Tiao Du Capsule 40mg/100g/ml,and the high-dose group were feed 80mg/100g/ml. 500g mash were given everyday each group and measured its remaining amount in the other day.We calculate the amount of liquid intake by giving each group 250ml water and 250ml sucrose solution one day and measuring its remaining amount in the other day.The amount of water intake=250- remaining water amount.The amount of sucrose solution intake=250- remaining sucrose solution amount.The hobbies ratio = The amount of sucrose solution intake/the amount of liquid intake.All the rats were prohibit eating for 24h after 7 days treatment.The difference of the substance p were measured by using Immunohistochemistry. After treatment, the experiment of emotional behavior were tested by open field reactiveness, resisting capture responsiveness, raised maze method for observing escape time in 5min from open Arm to surround Arm and enter times,and Y-maze for observing the frequency of errors.Results: 1, The food-intake difference of rats among groups:the food intake of the normal group was stable.In the 7 days stimulation,the food intake of model groups were decreasing obviously compared with the normal group.After the medical treatment,the food intake of the chai hu shu gan group, deanxit group,low-dose and high-dose group were increasing obviously,there is no significant difference compared with the normal group.2, The amount of liquid intake and hobby ratio:compared with normal group ,the amount of liquid intake in the model group was decreasing obviously, there is a significant difference(p﹤0.05).The difference of hobby ratio between the normal group and model group was obvious(p﹤0.05).3. The activity points of the rats in different groups:compared with normal group,the points of the model group was increasing obviously ,there is a significant difference(p﹤0.05). There is a significant difference between the medical treatment group and model group(p﹤0.01).4.Theresult of Boosted Maze experiment:compared with the normal group,the lasting time in the Open arms of the model group was increasing obviously, there is a significant difference(p﹤0.05). There is a significant difference between the medical treatment group and model group(p﹤0.01).compared with normal group,the entering frequency to the open arms of the model group was increasing obviously, there is a significant difference(p﹤0.05). compared with model group, The deanxit group and Tian Bing Tiao Du group was decreasing obviously, there is a significant difference(p﹤0.05).5.The result of Y-maze test: compared with the normal group,error times of selecting in the model group on the first day and second day was increasing obviously, there is a significant difference(p﹤0.05),the medical treatment groups were decreasing obviously compared with the model group, there is a significant difference(p﹤0.01).6. The result of positive products of substance p:The positive products in the gastric antral of the model group was decreasing obviously compared with the normal group, here is a significant difference(p﹤0.01).The medical treatment groups were increasing obviously, there is a significant difference compared with the model group (p﹤0.05).Seeing from the means,the high-dose group was the highest,it was 84.51±11.26. The positive products in the rats'Hippocampal ca1 area and Ventral hypothalamic nucleus was decreasing obviously compared with the normal group, there is a significant difference(p﹤0.05). The medical treatment groups were increasing obviously, there is a significant difference compared with the model group (p﹤0.01).Seeing from the means,the high-dose group was the highest,it was 34.68±8.76 and 18.78±4.23.Conclusion:1. There are significantdifference of the amout of the food-intake, the emotional behavior in the model group rats compared with the normal group, which is similar to the Disharmony of liver and stomach in the traditional Chinese medicine.2.There is a significant decreasing of substance p in the rats'gastric antral and brain of the model group. 3 Tianbing Tiaodu Capsule can improve food-intake, appetite, and mood of the irritation and stress rats,which were as clinical medication evidence.4. Tianbing Tiaodu Capsule could improve the substance p , which were relate to adjust Abnormal feeling of the irritation and stress rats.
