Depression is a mental disorder, or emotional disorder, it is a common disorder. Clinical study found that patients with depression have not only mood symptoms but also some physical symptoms, such as irritable bowel syndrome,functional dyspepsia; Depression with severe body discomfort Influence people's health. Therefore, to treat depression with IBS or FD, the research is focused on the pathogenesis of depression and treatment of gastrointestinal discomfort. Chinese medicine--bulbus lilium has a long history treatment of consciousness disorders, It can not only treat cough due to deficiency of the lung, lung abscess, deficient dysphoria and terror, staring spells, insomnia and dreaminess and so on, it can also be used in fullness, abdominal distention and other treatment. A number of clinical studies and the basis of previous studies have shown that the total saponins in bulbus lilii has a role of antidepressant.In this study, based on previous work, the total saponins in bulbus lilii of quality standards and Optimization of extraction and purification were studied, and the three Pharmacopoeia efficacy antidepressant bulbus lilii were compared to study the better efficacy of a further depth study of the depression and improvement of gastrointestinal symptoms.Aimed at establishing the criteria of quality control of total sponins extract in bulbus lilii to effectively control the quality of the effective parts and study their effects on the central nervous system and gastrointestinal system. It provides a theoretical basis for the fully development of bulbus lilii.Studies were carried on the extraction and purification process of total saponins by reviewing the impact of ethanol concentration and times. the best extraction process as 10BV 70% ethanol distilling two times,3h per time. Extract of total saponins was purified with macroeticular resins. It was confirmed that the best purification process was AB-8 macroeticular resins, medical material dried resins being 2:1, sample fluid concentration being 0.10g dried medicinal herb per milliliter, eluted by 6BV 80% ethanol after 6BV water and 5BV 30% ethanol with flow rate of 2ml/min.80% ethanol part was collected and dried under vacuum resulting in the total saponins content reaching 57%.Total saponins extract were prepared by 10 lots bulbus lilii. The studies established TLC method, confirmed the limits of loss on drying, established quantitative analysis method of total saponins by ultra-voilet spectropho-tometry and dioscin by RP-HPLC-ELSD for establishing the criteria of quality control of bulbus lilii total saponins extract.The antidepressant efficacy of three lily was comparative studied by the tail suspension test in mice, reserpine antagonistic test and swimming test; it obtained that lancifolium is better; and lancifolium was further studied in the following antidepressant study. By Using 5-HTP-induced behavior response and and yohimbine caused death in mice experiments, the total saponins enhances in mice shakes, but no enhancement of the role of yohimbine toxicity, It suggest that its antidepressant activity may be related to increased brain 5-HT nerve function, whereas the NE nerve function may be independent.The chronic unpredictable stress model was set up. The percentage of sucrose consumption and the immobitlity time in FST were observed to assess the model. The brain-gut peptides were detected in blood, stomach, intestines and other tissues. The results show that total saponins can increase VIP plasma levels, decrease in VIP content in colon tissue, and increase serum and colon tissue GAS, SP and other content.the total saponins above study shows the intervention can be direct or indirect function of the central nervous system of the digestive tract. Pharmacological tests show that total saponins of bulbus lilii can improve depression symptoms and gastrointestinal symptoms, which may be refer to the brain-gut axis and 5-HT nervous system, but what is its exact mechanism, remains to be further studied.
