| Purpose:Through exploring the influence of the traditional Chinese medicine Tong Guan Fang on the expression of EGFR and ICAM-1 about the tubal epithelium cells of FTIOI rat models to provide the experimental basis for the clinical application.Method:Choosing 180 Wistar female rats as the subjects and randomly extracting 30 female rats as the normal group, later using the mixed bacteria inoculation method to establish FTIOI rat models; After making modles success, rat models will be randomly divided into model group for 24 rats, therapeutic group for 25 rats, Tong Guan Fang high, medium and low dose group for every 24 rats. Each group is divided into two time groups for 15 days and 30 days; observing the change of the oviduct organization morphology under HE dyeing, and using the immunohistochemical method to observe the tubal epithelium cells EGFR and ICAM-1 protein's expression of all groups of rats and the conception of rats.Results:1. The observation to the tubal organization morphology of rat models is in accord with chronic inflammation's performance under HE dyeing, it causes infertile. Through the treatment, the condition of all groups make varying degrees of progress, and the conception rate increases, but Tong Guan Fang high-dosage group has the most significant improvement (P< 0.05).2. EGFR protein has a small amount of positive expression in the normal rats's tubal epithelium cells, EGFR protein's expression obviously enhances in the model group. Compared with the normal group, the model group has significant statistical significance(P<0.01); The expression of Tong Guan Fang high-dosage group in 15 days and 30 days are obviously below to the model group, the therapeutic group, the low-dosage group, the medium-dosage group (P<0.05 or P<0.01); The expression of Tong Guan Fang medium-dosage group in 15 days and 30 days are obviously below to the model group (P<0.05 or P<0.01), it also has difference from the therapeutic group in 30 days (P<0.05); compared with the model group in 30 days, Tong Guan Fang low-dosage group has significant statistical difference (P<0.01); but compared with the therapeutic group, they have not statistical difference(P> 0.05).3. ICAM-1 protein has not expression in normal group, but in the model group it exists high expression, so the comparative differences have remarkable statistical significance between the two groups (P< 0.01). The high-dosage group's expression in 15 days and 30 days are obviously below to the model group, the therapeutic group, the low-dosage group and the medium-dosage group (P<0.01); The medium-dosage group's expression in 15 days and 30 days are obviously below to the model group (P<0.05 or P<0.01), and compared with the therapeutic group, the difference of expression in 30 days also has statistical significance(P <0.05); The low-dosage group in 30 days is obviously below to the model group (P<0.01), but compared with the therapeutic group, it has not statistical difference(P>0.05).Conclusions:1. FTIOI rat models can be successfully copied by using mixed bacteria inoculation method.2. Tong Guan Fang can improve the structure and function of the fallopian tube of FTIOI rat models, increase the conception rate.3. Tong Guan Fang can inhibit the overexpression of the tubal epithelium cells EGFR protein and the expression ICAM-1 protein, thus plays its anti-inflammatory effect, it may be one of the mechanism of treating FTIOI. |
