Objective:By experiment to observe effect of the hirudin through pelvic cavity perfusion on the tubal pathological changes and expression of Bcl-2、EGFR of the fallopian tube inflammatory obstructive infertility rat models,to explore its action mechanism,providing a new method and theoretical basis for treatment of oviduct inflammatory infertility.Methods:Using a clean health 100 unfertilized SD female rats,randomly divided into normal group, the remaining 80 rats with mixed bacteria inoculation injection copy rats,s fallopian tube inflammatory obstructive model.Building the model 20 days later the remaining 65(only for death in the process of building 11 and anatomic 4)were randomly divided into three groups, namely model group(21 rats), frangrant ainsliaea herb group(22rats), hirudin group(22 rats), under the same conditions, molding 20 days later began to pelvic cavity perfusion, pelvic cavity perfusion the equal amount of normal saline into normal group and model group.Each group was further divided into two time groups(15 days, 30days)which were treated for 15 days,30 days respectively,at two different time points killing them(6-9only), and takeing the fallopian tubes. Macroscopic observation the general form and changes of the fallopian tubes. HE dyeing observation tubal histomorphology changes, Using immunohistochemical detection the tubal epithelium cells Bcl-2 、 EGFR protein‘s expression.Observe remaining rats each groups of 4 to 5 only which had stopped treating was pregnant situation.Results:1.The fallopian tube inflammatory obstructive model was established by injecting the mixed bacteria. After building the model 5, 10, 15, 20 days,selected rat oviduct tissue randomly. Through Macroscopic observation and HE staining observation to confirm copy the successful model which after building 20 days.2.To observe rat oviduct after treatment for pelvic cavity perfusion,Comparedwith model group, Hirudin group and frangrant ainsliaea herb group to varying degrees to improve model oviduct histomorphology in rats.3.The expression of BcL–2 in rats oviduct epithelium cells:In normal group rats the BcL–2 is not expressed in oviduct epithelial cells,and expressed in model group significantly. Two groups have significant difference is statistically significant(P<0.01). Compared with model group, frangrant ainsliaea herb group and hirudin group are different degree of inhibition of the expression of BcL-2 in the model rats,s oviduct epithelial cells(P < 0.05).4.The expression of EGFR in rats oviduct epithelium cells:In normal group rats the EGFR has a small amount of positive expression in oviduct epithelial cells,but expressed in model group significantly increased. Two groups have significant difference is statistically significant(P<0.01). Compared with model group, frangrant ainsliaea herb group and hirudin group are different degree of inhibition of the expression of EGFR in the model rats,s oviduct epithelial cells(P < 0.05).5.Treatment with 15 days compare with treatment 30 days : ① The normal group, model group of Bc L-2 showed no significant difference(P>0.05); There are differences in hirudin group and frangrant ainsliaea herb group(P<0.05); ② The normal group, model group of EGFR showed no significant difference(P>0.05);There are differences in hirudin group and frangrant ainsliaea herb group(P<0.05).6.Observe the conception of rats after pelvic cavity perfusion treatment 30 days,model group conception rate is 20%, lower than normal group significantly, each treatment group conception rate increased after treatment.Hirudin group compared with frangrant ainsliaea herb groups has no statistical significance(P > 0.05).Conclusion:1.The tubal inflammatory obstruction model can be established successfully by injecting the mixed bacteria.2.Hirudin though pelvic cavity perfusion can significantly reduce rat oviduct inflammatory, improve the probability of conception; The effect after being treated by hirudin for 30 d is better than that for 15 d.3.Hirudin through pelvic cavity perfusion can inhibit the expression of BcL-2protein and EGFR protein in the fallopian tube epithelial cells of model rats, promotethe inflammation cell and other hyperplasia of cells apoptosis, regulating cell growth, promote restoration of the damaged tissues, so as to give play to the role of anti-inflammatory, this may be one of the mechanism of action of fallopian tube inflammatory obstructive infertility treatment.
