Part one:Establishment of the seizures induced by electrical stimulation of rat dura mater modelObjective:To determine the convulsion threshold by electrical stimulation of rat cerebral dura mater and observed the changes in behavior of the rats after the stimulations.Methods:The 30 rats were randomly divided into operation group, intervention group and sham surgery group. In this study determination of the threshold for convulsions is made by direct cerebral dura mater electrical stimulation with different current intensity in rats. We measured the convulsive threshold and observed behavioral change in rats.Results:All rats occurred convulsion when the current reaches a certain intensity of stimulation. Operation group and the seizure of drug intervention group achieved gradeâ… -â…¡and gradeâ…¢-â…£of the current intensity was significantly different (P<0.05); Operation group and intervention group compared with the sham group significant behavioral changes (P<0.05), but the operation group and intervention group no significant difference (P>0.05).Conclusions:The seizures induced by electrical stimulation of the dura is a relatively simple and safe preparation of the animal model, application of antiepileptic drugs can increase seizure threshold, which provides a convenient model for investigating anticonvulsant drug effects. Seizure duration and intensity can be controlled, it is suitable to study the different time of the seizure expression of neuroglobin ideal animal model. Part two:The expression of neuroglobin in the preliminary study under different time of seizureObjective:To study the expression of neuroglobin in convulsion under different timeMethods:The 30 rats were randomly divided into sham operation group,5min group,15min group,30 minutes group and the delay group. Each group underwent HE staining and immunohistochemical staining, morphological changes were observed, compared at different times of the NGB's expression changes.Results:Compared with the sham group, other seizure groups significantly increased the expression of NGB, Compared among the groups,5min and 15min group compared with no significant difference (P>0.05); 15min group compared with the 30min group difference was significant (P<=0.05), compared with the rest of the group were significantly different (P<0.01).Conclusions:NGB expression increased rapidly after the seizures. NGB in the protection of hypoxic brain injury caused during seizures plays an important role, and this protection has a certain continuity.
