Objective1. To study PECAM-1 gene polymorphisms in the exons3 and 8 with coronary heart disease(CHD)among the Han nationality in the west of Liaoning. To explore the PECAM-1 gene polymorphism with degree of the coronary artery stenosis.2. To study the relationship between plasma levels of soluble PCAM-1 and coronary heart disease in the Han of Western Liaoning.MethodIn the study,153 subjects with CHD and 200 controls were enrolled in Han of Western Liaoning. Genomic DNA of peripheral blood was extracted by Takara DNA blood extraction kit. According to genetic sequence polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP),The PECAM-1 +~G373C and +A1688G were genotyped. Experimental results were analyzed by SPSS software. The relationship between genotype and coronary heart disease was tested by Logistic regression analysis. Plasma concentration of sPECAM-1 was measured by Enzyme-linked immunosorbent (ELISA). Automated Biochemieal Analyzer was used to measure the serum.Results1. There was significant different distributions of genotype and allele in PECAM-1 +G373C and +A1688G polymorphisms between CHD and control groups.2. Coronary artery stenosis in CHD patients with different levels of PECAM-1 + C373G gene distribution are studied, the risk of GC and GG genotype in patients with severe coronary stenosis was 2.49 and 2.95 times than CC genotype.3. A1688G was no significant differences in gene distribution with different degrees of coronary artery stenosis among the CHD.4. There was significant different in levels of serum PECAM-1 among CHD and control groups.5. PECAM-1 + G373C and + A1688G genotype and serum levels were significantly different in the CAD group.Conclusion1. In western liaoning Han population, PECAM-1+G373C and +A1688G gene polymorphisms are associated with CHD.2. PECAM-1 + G373C polymorphism genotypes and coronary artery stenosis were significant differences in the distribution .The risk of GC,GG carriers suffering from serious coronary artery stenosis may increase.3. PECAM-1 serum levels can be affected by both PECAM-1 +G373C and + A1688G polymorphism.
