PURPOSE: To detect the concentration of Heat Shock Protein 70 and metallothioneinsâ…¡' s expression in Rat lens after exposed to different level of mercury vapor, and discuss the role they played in the process of mercurialentis. METHODS: 56 SD rats were randomly divided into three groups:low Hg-exposed level group (n=24), high Hg-exposed level group(n=24) and control groups(n=8). Rats of Hg-exposed groups were inhaled Hg vapor 30mins by day,6 days a week, with 3mg/m3 in high level and 0.01mg/m3 in low level. Rats of control groups were executed at the first day and lens were isolated separately preserved by liquid nitrogen and 4% paraformaldehyde. At the end of 1st week,2nd week,3rd week,4th week after mercury vapor inhaled, lens were isolated separately preserved by liquid nitrogen and 4% paraformaldehyde. Immunohist-ochemical method and Western Blotting method were used to detect the expression of HSP70 and MT-â…¡in experimental and control lens. RESULTS: 1.The formation of mercurialentis in rat lens. Observe in different period after exposed to mercury vapor: the lens of the control group were completely clarity during experiment, the lens of low level Hg inhaled group began to partial anterior capsular opacity by week 3 and become punctiform anterior capsular opacity by week 4, the lens of high level Hg inhaled group began punctiform anterior capsular opacity by week 3 and accompany with cortical opacity.2. Immunohistochemical method and Western Blotting method indicate that the expression of HSP70 were significantly different between experimental and control LECs (P<0.05), and also significant different between low level group and high level group, the expression of HSP70 are correlated with density and time scale. The expression of MT-â…¡were significantly different between experimental and control LECs (P<0.05), and also significant different between time scale, but not significant correlated with density. The expression of HSP70 is positive related with MT-â…¡. CONCLUSIONS:The expression of HSP70 and MT-II are continually increased during exposed to mercury, and the expression of HSP70 are positive related with MT-â…¡, which indicated that the two protein played a critical role during the development of metallothioneins.
