Objective: As a active compound of GTF (glucose tolerance factor), chromium is an essential trace element to human and animals. Schwarz and Mertz found GTF, a substance which can regain the damage of rats endogenous glucose tolerance from pig kidney powder in 1957. Study found that brewers' yeast contains GTF, in whose active ingredients is trivalent chromium. Thus the people stablished the relationship with GTF and chromium. GTF is the most abundant in the yeast. Rich chromium yeast not only contain high chromium volume, but also contain rich protein, nucleic acid, glycogen, fat, biotin, a variety of vitamins B, cholesterol and other kinds of trace elements, therefore it has high nutritional value which attract more and more people's attention. In recent years, due to the improvement of living standards, dietary structure change and an increasingly tense rhythm of life, the incidence of diabetes is growing rapidly in our country, diabetes has become the third big serious threat to the health of the population of chronic diseases less than the tumor and cardiovascular disease. This thesis's research object is s.cerevisiae. The rich chromium yeast cultivation process was studied, and based on it, apply rich chromium yeast to type 2 diabetes rat models, through the blood glucose, blood fat, etc. biochemical changes before and after the experiment rats, confirmed the intervention role of rich chromium yeast to type 2 diabetes.Methods:1 Screening saccharomyces cerevisiae 2.0420 which can enrich and transform inorganic chromium to domesticate, adapt it to continuously heightened inorganic chromium salts environment; choose the yeast which have good adaptability, good enrichment capacity, high conversion speed to proliferate, obtain the bioactive chrome. 2 Orthogonal experimental design and determine the best combination of various components.3 (1) Observe the products'effect to blood glucose and blood lipid in type 2 diabetes rats, determine the adjusting effects to blood sugar and blood lipid in experimental animals. (2) Observe the products'effect to fat rats model weight, determine the inhibited obesity effects to experimental animals.Results:1 With s.cerevisiae 2.0420 for starting strains, wave bottle in the level of seeds medium is optimized, get the optimal seed medium: glucose 2.0%, urea 1.0%, yeast grease 1.0% ,chromium 50μg/mL, maLt juice 0.4% ph5.0. Compared with the original seed medium, organic chromium yield in fermentation liquor increased 41%, production for 10.87 mg/mL.2 The single factor experiment determine fermentation glucose concentration in medium for 4%, Urea concentration for 0.5%, The optimal pH is 5.0.3 Based on the study of fermentation of various factors , conducted orthogonal test, inspects the concentration of trace element to the influence of yeast organic chromium yield in fermentation medium, determine the sodium selenite 20μg/mL,sodium chloride 150μg/ml, sodium dihydrogen phosphate 0.5μg/mL, niacin 0.004μg/mL for optimal concentration, in these conditions, rich chromium yeast biomass for 9.07 mg/mL, organic chrome content for 2177μg/g, fermented liquid organic chromium yield for 19.87μg/mL.4 Irrigate to type 2 diabetic rats, irrigation type 2 diabetic rats stomach, obesity rat model group, high compared low dose rich chromium yeast rats, blood sugar significantly (P < 0.01), but higher than normal control group.5 Different degree of decline with rats blood lipid index appear both in high–dose and low-dose rich chromium yeast treatment groups.6 Rats body qualities in high–dose rich chromium yeast treatment group and low-dose rich chromium yeast treatment group are significantly lower than obesity model group.Conclusion: 1 Adjust glucose concentrations, improve medium components of urea can greatly improve concentration and GTF production.2 Rich chromium yeast have a role of preventing incidence of type 2 diabetes and reducing the level of blood sugar in type 2 diabetic rats.3 Rich chromium yeast have a role of adjusting the level of blood lipid in type 2 diabetic rats.4 Rich chromium yeast propolis mixture has inhibitory of type 2 diabetic rats weight over-increase role. Rich chromium yeast have a role ofinhibiting weight too fast growth in type 2 diabetic rats.
