Objective:To investigate the expressions of inducibe nitric oxide synthase(iNOS),transforming growth factor-β1 ( TGF-β1 ) and CD105 in primary pterygia,recurrent pterygia and in normal conjunctiva tissues.And to explore their role in the occurrence and development of pterygium.Methods:The 52 human pterygial tissue samples were obtained from the Department of Ophthalmology in the NO.1 Hospital of Hebei Medical University during 2009 to 2010,including 33 cases of primary pterygium,19 cases of recurrent pterygium.Specimens of normal conjunctiva were acquired from the surgons in the same period.Cryopreserved tissue specimens consisting of primary pterygia,recurrent pterygia and normal conjunctiva were used to study the expression of iNOS,TGF-β1 and CD105,using immunohistochemical S-P method.And the CD105-MVD was also determined in this study.Results:The positive rate of iNOS in primary pterygia,recurrent pterygia and normal conjunctiva were 84.8%,89.5% and 14.3%.Comparing the positive rate of iNOS between the primary pterygia and normal conjunctiva and between the recurrent pterygia and recurrent pterygia,they all had significant difference(p<0.001).Comparing the positive rate of iNOS between the primary pterygia and the recurrent pterygia, p>0.05,had no statistical meaning. The positive rate of TGF-β1 in primary pterygia,recurrent pterygia and normal conjunctiva were 63.6%,78.9% and 21.4%. Comparing the positive rate of TGF-β1 between the primary pterygia and normal conjunctiva and between the recurrent pterygia and recurrent pterygia,they all had significant difference(p<0.05). Comparing the positive rate of TGF-β1 between the primary pterygia and the recurrent pterygia, p>0.05,had no statistical meaning.The positive rate of CD105 in primary pterygia,recurrent pterygia and normal conjunctiva were 87.9%,94.7% and 28.6%. Comparing the positive rate of CD105 between the primary pterygia and normal conjunctiva and between the recurrent pterygia and recurrent pterygia,they all had significant difference(p<0.001).Comparing the positive rate of CD105 between the primary pterygia and the recurrent pterygia, p>0.05,had no statistical meaning.The MVD in the pterygia and normal conjunctive were 10.40±4.56 and 1.43±1.02,there was a significant difference between them(p<0.001).The correlation between the expression of TGF-β1 and CD105-MVD in pterygia was analyzed using Spearman rank correlation analysis,and they were turned out to be positively related.Conclusions:There were high iNOS,TGF-β1 and CD105 expression rates in pterygia,and the relationship between TGF-β1 and CD105 in pterygia was so close.All of these suggested that there was abnormal blood vessel multiply in pterytium,and the expression of iNOS,TGF-β1 and CD105 in pterygia is significantly associated with the clinic pathologic stage and recurrence.The specific iNOS,TGF-β1 and CD105 inhibitor may become a new target for the therapy of pterygium and reduce the postoperation recrudescence.
