Objective:To observe the effect to type 2 diabetic rats on serum levels of TNF-a, inquire into the mechanism of the ShenQi Compound's effect on protecting from diabetes and diabetic nephropathy,for rational use of the Compound for clinical drug development and to provide theoretical and experimental basis.Methods:GK rats were 16 weeks of age, while feeding high-fat diet for replication model of spontaneous type 2 diabetic nephropathy. Selected rats were randomly divided into four groups as ramipril group, Shen Qi compound recipe-high dosage group, ShenQi compound recipe-low dosage group,model group. Simultaneously 10 of 16 weeks normal SD rats (SPF) were set as the normal control group.ramipril group:by 1mg/(kg.d) fed with ramipril suspension; ShenQi compound recipe-high dosage group:by 2.88g/(kg.d) (equivalent to 20 times the adult dose, that is 4 times the human equivalent dose for adults at 60 kg total) fed with the Shen Qi Compound suspension; ShenQi compound recipe-low dosage group:by 0.72g/(kg.d) (equivalent to 5 times the adult dose, the human equivalent dose for adults at 60 kg total)fed with the ShenQi Compound suspension; Model group and normal control group:by 5ml (kg.d) fed with sterile water. Meanwhile, the normal control group fed normal diet, the other groups fed with high fat diet (normal diet 88.2%,10% of refined lard, cholesterol 1.5%,0.3% porcine bile salt), Observe the general condition during the experiment, water intake, food intake, body weight; After 8 weeks of continuous administration animals were sacrificed,12h fasting before death can not help but water,Sugar oxidase in Portugal rat tail vein blood glucose measured after the body weight by intraperitoneal injection of 8% 2ml/kg amobarbital sodium anesthetized animals, Abdominal aortic blood serum was separated,-20℃preservation, Measured by enzyme-coupled assay of cholesterol and triglycerides, renal function measured by oxidase method, enzyme-linked immunosorbent assay (ELISA), method to measure IL-6, TNF-α; The rat right kidney on ice, remove the envelope, take part of the renal cortex immersed in 2.5% glutaraldehyde phosphate buffer (pH= 7.2) in the fixed electron microscopy sections made to observe the ultrastructural changes of the kidney.Results:In ShenQi compound recipe-high dosage group the general state,weight gain, food intake and water intake of GK rats,was significantly better than the indicators in model group; Shen Qi compound can reduce the blood glucose, blood lipids, IL-6 and TNF-αlevels of GK rats,improve the renal function, compared with the model group were significantly different (P<0.05);Obviously, more pathological changes in the model group than the Shen Qi compound recipe-high dosage group.Conclusion:the Shen Qi Compound can significantly reduce the spontaneous type 2 diabetic GK rats with blood glucose, blood lipids, IL-6 and TNF-αlevel,delay the deterioration of renal function in Spontaneous 2DM GK rats,this may be one of the mechanisms to prevention from diabetes and diabetic nephropathy.
