Objective To research the mechanisms of ShenQi compound recipe (SQCR) treating type 2 diabetes mellitus (T2DM) on macrovascular complications and preserving vessel endothelium. To provid the theory bases of representing with SQCR applying Traditional Chinese Medicine in clinic. Methods copied T2DM model with macrovascular complications via injecting Nω-nitio-L-arginine methyl ester (L-NAME) intra-peritonelly 2 weeks and gavaging high fat diet 4 weeks. Divided animals into five groups according to the level of blood sugar: model, ramipril(western medicine), ShenQi compound recipe low dosage and high dosage (Traditional Chinese Medicine low dosage and high dosage), and normal control group and administrated water, ramipril, SQCR low dosage, SQCR high dosage, water respectively for 4 weeks after intra-peritonelly L-NAME 2 weeks except normal control group. During the experiment period, observed general status and measured the values of drinking, eating, body weight. At the end, estimated glycation serum protein (GSP) with Fructosamine assay; insulin (INS), interleukin-6 (IL-6), Endothelin-1(ET-1) with radio immunoassay; Nitric oxide(NO), nitric oxide synthase(NOS) with chemical shade selection; Oxidated low density lipoprotein (ox-LDL), intercellular adhesion molecule-1 (ICAM-1), with enzyme-linked immunosorbent assay (ELISA); thoracic aorta ICAM-lmRNA and CD36mRNA with real time RT-PCR; thoracic aorta pathomorphology with light microscope. Compared the correlation with NO and ET-1, ox-LDL and CD36mRNA. Results Values of drinking weight of SQCR low dosage and high dosage group were decreased remarkable (p<0.01).To compared with model group, the body weight of SQCR low dosage and high dosage group gained remarkable (p<0.01). Content of IL-6, ET-1, INS, GSP, ICAM-1, ox-LDL were all decreased in SQCR low dosage and high dosage group compared with model group (p<0.01 or p<0.05). Content of NO and NOS were increased in SQCR low dosage and high dosage group compared with model group (p<0.01). The expression of thoracic aorta ICAM-lmRNA and CD36mRNA in SQCR low dosage and high dosage group were also decreased remarkable compared with model group (p<0.01). NO and ET-1 had correlated negatively with lined (r=-0.90) , ox-LDL and CD36mRNA's Ct had correlated negatively with lined (r=-0.60).
