Modern pharmacology study shows that coptis have pharmacological effects such as antipyretic, antibacterial, antitumor, arrhythmia, fall blood sugar etc., which is mainly produced in Chongqing, Sichuan, Hubei, etc. Cortex Moutan, mainly produced in Dianjiang, Bozhou city, Heze city, Luoyang city, etc., contains the many effective ingredients such as paeonol, which are the important basis of cortex moutan quality. Good efficacy needs excellent quality guarantee, it is the key that the evaluation and control quality of herbal medicine. This experiment is established HPLC fingerprint of coptis and moutan, the various chemical compositions contents were determined by HPLC. The relationship between various chemical compositions and environment factors, was preliminary established a chemical identification method for coptis and moutan. It is provided that scientific basis for the quality control of coptis and moutan.Objective:Study on quality of coptis and moutan by establishing HPLC fingerprint and determining its contents of various chemical compositions.Method:1. Collect samples of different producing areas, separate and purify various chemical compositions.2. Establish HPLC fingerprint and determine the contents of various chemical compositions in coptis and moutan.3. Determine the contents of aflatoxin by TLC, and the contents of heavy metal by Atomic absorption spectrophotometry.4. The differences of coptis and moutan, among various origin, chemical compositions contents and environment factors were evaluated by mathematical statistics such as clustering analysis, similarity analysis, gray relation, Q inspection and G inspection, etc.Result:1. Collect 119 coptis and 156 moutan from different producing areas. Establish the HPLC method, the average recovery at 97-103%, RSD≤3%, for determination various chemical constituent contents.2. 27 coptis and 46 moutan samples were detected by HPLC fingerprint,12 common peaks from coptis and 13 common peaks from have moutan had been calibrated. The cluster analysis and similarity analysis results show that the chemical constituents of different areas have good consistency.3. There are good relationship between chemical composition and environmental factors. Combined with the content determination results, the quality of coptis is Enshi> Zhuxi> Emei> Shizhu> Lichuan, the quality of moutan is Nanlin> Bozhou> Tongling> Heze> Luoyang> Caoxian> Dianjiang.4. The result of determination aflatoxin showed that it's a good job to control fungi tumor. The result of determination heavy metal showed that the content exceeds standard especially Cu.Conclusion:1. The method of quantitative determination of six alkaloids in coptis and five chemical compositions in moutan, which is efficiently, quickly and accurately, was provided the chemical basis for quality control of coptis and moutan.2. The correlation can provide scientific proof for coptis and moutan later cultivation between chemical composition and environmental factors. Established method of quality evaluation reflecting its genuineness of coptis and moutan can provide reference for other authentic herbs.3. Because of the contents of heavy metal were exceed standard, It must be strengthened that environmental monitoring in medicinal material production base.
