This dissertation consists of three parts:Chapter I Summary of the related literaturesPoria cocos, also named Fuling, is the dried sclerotia of Poria cocos (Schw.) Wolf, belonging to PolyPoraceae. In this part, the related literatures about the chemical constituents, pharmacological activities of Poria cocos and quality conctrol were reviewed on the basis of138references. Polysaccharides and triterpenoids are the main constituents of Poria cocos and24and83compounds have been reported up to now, respectively. It also has a wide range of pharmacological activities, such as anti-tumor, regulation of immune function, anti-inflammatory, anti-aging. The reaserches on quality control mostly focused on determination and fringerprints of active constituents.Chapter II Studies on chemical constituents of Poria cocos (Schw.) WolfThe researches about chemical constituents of Poria cocos (Schw.) Wolf have been carried out by various types of chromatography and spectroscopy, including TLC, Sephadex LH-20, MCI, HPLC, NMR, and so on. Four compounds were obtained and identified. They are hesperidin, protocatechuic acid, pregn-7-ene-2β,3a,15a,20(S)-tetrol, phenylalanine. Among them, hesperidin, protocatechuic acid and phenylalanine were firstly found from Wolfiporia.Chapter Ⅲ Studies on quality control methods for Poria cocos (Schw.) WolfThe HPLC fingerprints of Poria cocos (Schw.) Wolf were established with the detection wavelength254nm and analyzed by using Chromatographic Fingerprint Evaluation System(A version). There were19common peaks in the fingerprints. The similarity analysis of15samples from different regions revealed that the similarities were between0.985and0.860with10regions above0.9and6regions above0.95. Clustering results showed that15batches of Poria cocos were clustered into three categories. In the principal component analysis, six principal components were extracted, and the accumulated contribution rate was91.647%, in which the first principal component contribution rate was31.869%. The peaks6,13,4,7,15contributed the most to the first principal component. The content of the5components may be the main reason for the clustering result.The qualitative identification of the common peaks of the Poria cocos fingerprints were carried out by using LCMS-IT-TOF.17compounds in the19common peaks were identified together with another3compounds with only end absorption. Common peaks, in some extent, are reflections of fingerprint informations and chemical constituents of medicinal materials. So it can provide a reliable basis for the quality control of Poria cocos (Schw.) Wolf from different regions by qualitative analysis of common peaks.A quantitative method was firstly established for the determination of Pachymic acid and Dehydroeburicoic acid by using High Performance Liquid Chromatography. The regression equations of the two compounds were Y=491402X+75835(r=0.9994) and Y=591020X-6847.4(r=0.9996), respectively. The linear range of Pachymic acid was0.424μg-3.392μg and Dehydroeburicoic acid0.0458μg-0.458μg. The average recoveries were101.85%with RSD of2.43%for Pachymic acid,100.95%with RSD of1.58%for Dehydroeburicoic acid, respectively. The content determination results of the two compounds in15batches of raw drugs showed that the methods were simple, quick, reliable reproducible and can be used as the determination of Poria cocos.Innovation:1. Hesperidin, protocatechuic acid and phenylalanine were obtained from Poria cocos (Schw.) Wolf for the first time, and were also firstly found from Wolfiporia..2. A quantitative method for the determination of Pachymic acid and Dehydroeburicoic acid was firstly established by using HPLC.
