Objective This study was designed to evaluate the skeletal effects of s-3-(4-acetyla mino-phenoxy)-2-hydroxy-2-methy1-N-(4-nitro-3-trifluoromethyl-phenyl)-propionamide in an osteopenic model.Methods Osteoporosis rats model was developed successfully by tcastrate, then treatment with subcutaneous injection of Dihydrotestosterone (DHT)and Sarm was applied in the next 12 weeks.Measurement of bone mineral density(BMD) and biomechanical properties of femur and lumbar vertebra,prostate weight and biochemical assays of serum were recorded after the termination of therapy.Result 1.The BMD of orx group in whole-body,femur and lumbar vertebrae is obviously lower than sham group(P<0.001),treatment using Sarm of middle and high is obviously higher than orx group(P<0.01),The BMD of treatment using Sarm of low increased BMD but not obviously.The BMD of treatment using Sarm of middle ,high and DHT group is obviously higher than sham group(P<0.01),The group of treatment using Sarm of middle ,high and DHT was showed no significant difference.2. The Biomechanical properties of orx group in femur and lumbar vertebrae is obviously lower than sham group(P<0.001), treatment using Sarm of middle and high is obviously higher than orx group(P<0.01),The group of treatment using Sarm of low increased Biomechanical properties but not obviously.The Biomechanical properties of treatment using Sarm of middle ,high and DHT group is obviously higher than sham group(P<0.01),The group of treatment using Sarm of middle ,high and DHT was showed no significant difference.3.the orx group serum Ca obviously decrease,serum P obviously increase ,serum BALP obviously increase (P<0.001);after 12 weeks treatment, serum Ca and serum P of every group was showed no significant change. The serum BALP of treatment using Sarm of middle ,high and DHT group is obviously;lower than orx group(P<0.001),The serum BALP of treatment using Sarm of middle ,high and DHT group is obviously lower than orx group(P<0.001),The group of treatment using Sarm of middle ,high and DHT was showed no significant difference.4.The prostate wet wight and prostate exponent of sham group is obviously higher than orx group(P<0.001), The group of treatment using Sarm of low,middle and sham was showed no significant difference. The group of treatment using Sarm of high prostate slightly hyperplasia.it was showed significant difference and Biomechanical properties,Meanwhile,it decreased the levels of serum Bone Alkaline phosphatase,but not to cause prostate significant hyperplasia(P<0.05);The groups of treatment using Sarm was showed no significant difference.,not to cause prostate significant hyperplasia.Conclusion The ability of Sarm to promote bone mineral density and Biomechanical properties of osteopenic male rats.Position these drugs as promising new alternatives for the treatment of osteoporosis.
